Pontibacillus chungwhensis gen. nov., sp. nov., a moderately halophilic Gram-positive bacterium from a solar saltern in Korea

Abstract: Three moderately halophilic, spore-forming strains, designated BH030062T, BH030049 and BH030080, were isolated from a solar saltern in Korea. Phylogenetic analyses and comparative 16S rRNA gene sequence studies revealed that the isolates represent a novel distinct monophyletic lineage within the phyletic group classically defined as the genus Bacillus and are most closely related to members of the genera Gracilibacillus (93·7–95·1 % similarity), Virgibacillus (93·5–94·8 %), Halobacillus (94·8–95·9 %), Filobaci… Show more

“…Flagella were stained according to the method of Leifson (Smibert & Krieg, 1981). Growth was tested at various temperatures (5-55 u C, in increments of 5 u C) on MA5 and at different (Lim et al, 2005), and on some other media as controls [MA, trypticase soy agar (Difco) and ISP medium 2 agar (Shirling & Gottlieb, 1966)]. Aesculin hydrolysis, nitrate reduction and H 2 S and indole production were determined as recommended by Smibert & Krieg (1994).…”

“…The genus Pontibacillus was first proposed by Lim et al (2005a). Members of this genus are Gram-positive, moderately halophilic, strictly aerobic, motile, endosporeforming rods, having a meso-diaminopimelic acid cell-wall type and containing MK-7 as the predominant respiratory quinone.…”

“…Members of this genus are Gram-positive, moderately halophilic, strictly aerobic, motile, endosporeforming rods, having a meso-diaminopimelic acid cell-wall type and containing MK-7 as the predominant respiratory quinone. At the time of writing, the genus comprises two recognized species, Pontibacillus chungwhensis (Lim et al, 2005a) and Pontibacillus marinus (Lim et al, 2005b).During an investigation of the diversity of the microbial population of invertebrates inhabiting the South China Sea, we isolated a moderately halophilic bacterium, designated JSM 076056 T , from a sea urchin collected from Leizhou Bay, China. On the basis of the results of a polyphasic taxonomic study, this strain represents a novel species of the genus Pontibacillus.…”

“…Moderately halophilic bacteria that grow optimally in media containing 3-15 % (w/v) NaCl are widely distributed in different marine environments and are made up of very heterogeneous physiological groups taxonomically, including both Gram-positive and Gram-negative microorganisms (Ventosa et al, 1998;Oren, 2002;Jeon et al, 2005a;Lim et al, 2005). Aerobic, spore-forming, halophilic, Gram-positive rods that were originally assigned to the genus Bacillus are also very diverse taxonomically and they form several phylogenetically distinct lineages (Ash et al, 1991;Stackebrandt & Liesack, 1993;Nielsen et al, 1994;Jeon et al, 2005b).…”
