Pontibacillus marinus sp. nov., a moderately halophilic bacterium from a solar saltern, and emended description of the genus Pontibacillus

Abstract: Pontibacillus marinus sp. nov., a moderately halophilic bacterium from a solar saltern, and emended description of the genus Pontibacillus A moderately halophilic, Gram-positive, rod-shaped bacterium (BH030004 T ) was isolated from a solar saltern in Korea. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain BH030004 T belonged to the genus Pontibacillus. Chemotaxonomic data (DNA G+C content, 42 mol%; major isoprenoid quinone, MK-7; cell-wall type, A1c-type meso-diaminopimelic acid; major… Show more

“…Acids are produced from D-mannose, D-ribose and starch, but not from Table 1. Differential characteristics for strain JSM 076056 T and type strains of recognized Pontibacillus species Data for JSM 076056 T are from the present study and those for P. chungwhensis BH030062 T and P. marinus BH030004 T are from Lim et al (2005a) and Lim et al (2005b), respectively. All strains are moderately halophilic aerobes comprising rodshaped, Gram-positive cells with swollen sporangia.…”

“…Members of this genus are Gram-positive, moderately halophilic, strictly aerobic, motile, endosporeforming rods, having a meso-diaminopimelic acid cell-wall type and containing MK-7 as the predominant respiratory quinone. At the time of writing, the genus comprises two recognized species, Pontibacillus chungwhensis (Lim et al, 2005a) and Pontibacillus marinus (Lim et al, 2005b).During an investigation of the diversity of the microbial population of invertebrates inhabiting the South China Sea, we isolated a moderately halophilic bacterium, designated JSM 076056 T , from a sea urchin collected from Leizhou Bay, China. On the basis of the results of a polyphasic taxonomic study, this strain represents a novel species of the genus Pontibacillus.…”

“…The topology of the phylogenetic tree was evaluated by the bootstrap resampling method of Felsenstein (1985), with 1000 replicates. The similarity values were calculated using MEGA3 (Kumar et al, 2004 Lee et al (2006), Heyndrickx et al (1998), Yoon et al (2002), Spring et al (1996), García et al (2005), Lim et al (2005a, b) and Wainø et al (1999). “…DNA-DNA hybridization experiments were carried out to evaluate levels of relatedness between strain EMB325 T and S. psychrophila LMG 5408 T and S. metamorpha DSM 1837 T . Extracted genomic DNA samples for slot hybridization were fragmented with restriction enzyme HaeIII, and random primed DNA labelling with digoxigenin-dUTP and hybridizations were performed using a digoxigenin High Prime DNA labelling kit (Roche Applied Science) according to the manufacturer's instructions and standard procedures (Sambrook & Russell, 2001;Lim et al, 2005). Phylogenetic analysis using the 16S rRNA gene sequence of strain EMB325 T showed that it formed a phylogenetic lineage with S. psychrophila LMG 5408 T and S. metamorpha DSM 1837 T ( Fig.…”
