Population-based approaches to genetics of migraine

Abstract: We argue that the detailed genetic architecture of migraine, various aspects of methodology, and the ultimate sample size achieved by population-based ascertainment will be critical determinants of the future success of this approach to genetic analysis of migraine and its comorbidities.

“…These public–private partnerships should be encouraged in future studies as they benefit all interested parties, and the pooling of all available resources can only help speed up new discoveries. Of course, it should be remembered that the contributions from academic or clinical institutions are equally important because these tend to consist of more severe cases and better phenotyped collections that can also improve power and enable more detailed follow-up questions to be investigated [5]. …”

“…65 However, population-based studies such as the CLSA have other strengths, including high statistical power given the large sample surveyed. Therefore, it should be noted that our population-based ascertainment of migraine is both less precise and likely more heterogeneous than if the gold standard of classification using specific diagnostic criteria were utilized.…”

“…Despite extensive research, its pathomechanism is still questionable. Migraine is heritable with estimated values of 0.34-0.57 (Chasman et al, 2016) reflecting an important role of environmental factors, also. A large number of genes of small effects appear to be involved in migraine (Anttila et al, 2013;Gormley et al, 2016), and their interactions with environmental factors have also been suggested (Sauro and Becker, 2009;Juhasz et al, 2017) but there is a significant lack of gene × environment interaction studies of migraine.…”
