Population structure of Japanese extraintestinal pathogenicEscherichia coliand its relationship with antimicrobial resistance

Abstract: More than half of the ExPEC population in Japan consisted of 11 major clones. Of these clones, the CH40-30-ST131-C/H30 clone was the predominant antimicrobial-resistant population. The presence of major clones with low-level ciprofloxacin resistance supports the potential future success of a non-ST131 fluoroquinolone-resistant clone.

“…Among the clade C isolates, the H30Rx status defined by the ybbW SNP-based PCR assay corresponded to subclade C2, except for 1 subclade C2 isolate in the ExPEC collection (BRG145, as mentioned above). These 86 C2 isolates were associated with bla CTX-M-15 (94.2%; whose presence was defined in previous studies [15,16]), confirming their relationship (4). The 202 C1-M27 and 180 C1-nM27 subclade isolates were associated with bla CTX-M-27 (98.0%) and bla CTX-M-14 (61.1%), as expected from our previous studies (8,15,16).…”

“…We investigated a total of 329 clinical E. coli isolates that were consecutively collected by a Japanese multicenter surveillance program (14). Of these isolates, 95 isolates (29%) were nonsusceptible (intermediate, 60 isolates; resistant, 35 isolates) to SAM, and 61 isolates (19%) were nonsusceptible to AMC.…”

“…2 , 12 Data from Japanese Nosocomial Infections Surveillance suggest that E.coli causing bacteremia is increasing in the Japanese population. 14 Since antibiotic resistant ExPEC strains are also prevalent in the Japanese population, 15 the Phase 1 study described herein was conducted to investigate the safety, tolerability and immunogenicity of 3 different dose levels of ExPEC4V vaccine in healthy adult Japanese participants. This study was conducted in a dose-ascending manner as there was no prior clinical experience in the Japanese population.…”
