ObjectiveIschemic stroke (IS) is a common disease that causes severe and long-term neurological disability in people worldwide. Although rehabilitation is indispensable to promote neurological recovery in ischemic stroke, it is limited to providing a timely and efficient reference for developing and adjusting treatment strategies because neurological assessment after stroke treatment is mostly performed using scales and imaging. Therefore, there is an urgent need to find biomarkers that can help us evaluate and optimize the treatment plan.MethodsWe used data-independent acquisition (DIA) technology to screen differentially expressed proteins (DEPs) before and after ischemic stroke rehabilitation treatment, and then performed Gene Ontology (GO) and pathway enrichment analysis of DEPs using bioinformatics tools such as KEGG pathway and Reactome. In addition, the protein–protein interaction (PPI) network and modularity analysis of DEPs were integrated to identify the hub proteins (genes) and hub signaling pathways for neurological recovery in ischemic stroke. PRM-targeted proteomics was also used to validate some of the screened proteins of interest.ResultsAnalyzing the serum protein expression profiles before and after rehabilitation, we identified 22 DEPs that were upregulated and downregulated each. Through GO and pathway enrichment analysis and subsequent PPI network analysis constructed using STRING data and subsequent Cytoscape MCODE analysis, we identified that complement-related pathways, lipoprotein-related functions and effects, thrombosis and hemostasis, coronavirus disease (COVID-19), and inflammatory and immune pathways are the major pathways involved in the improvement of neurological function after stroke rehabilitation.ConclusionComplement-related pathways, lipoprotein-related functions and effects, thrombosis and hemostasis, coronavirus disease (COVID-19), and inflammation and immunity pathways are not only key pathways in the pathogenesis of ischemic stroke but also the main pathways of action of rehabilitation therapy. In addition, IGHA1, LRG1, IGHV3-64D, and CP are upregulated in patients with ischemic stroke and downregulated after rehabilitation, which may be used as biomarkers to monitor neurological impairment and recovery after stroke.

Aim: To determine the effects of astaxanthin treatment on lipids, cardiovascular disease (CVD) markers, glucose tolerance, insulin action and inflammation in individuals with prediabetes and dyslipidaemia.Materials and Methods: Adult participants with dyslipidaemia and prediabetes (n = 34) underwent baseline blood draw, an oral glucose tolerance test and a onestep hyperinsulinaemic-euglycaemic clamp. They were then randomized (n = 22 treated, 12 placebo) to receive astaxanthin 12 mg daily or placebo for 24 weeks. Baseline studies were repeated after 12 and 24 weeks of therapy.Results: After 24 weeks, astaxanthin treatment significantly decreased low-density lipoprotein (À0.33 ± 0.11 mM) and total cholesterol (À0.30 ± 0.14 mM) (both P < .05). Astaxanthin also reduced levels of the CVD risk markers fibrinogen (À473 ± 210 ng/mL), L-selectin (À0.08 ± 0.03 ng/mL) and fetuin-A (À10.3 ± 3.6 ng/mL) (all P < .05). While the effects of astaxanthin treatment did not reach statistical significance, there were trends toward improvements in the primary outcome measure, insulin-stimulated, whole-body glucose disposal (+0.52 ± 0.37 mg/m 2 /min, P = .078), as well as fasting [insulin] (À5.6 ± 8.4 pM, P = .097) and HOMA2-IR (À0.31 ± 0.16, P = .060), suggesting improved insulin action. No consistent significant differences from baseline were observed for any of these outcomes in the placebo group. Astaxanthin was safe and well tolerated with no clinically significant adverse events.Conclusions: Although the primary endpoint did not meet the prespecified significance level, these data suggest that astaxanthin is a safe over-the-counter supplement that improves lipid profiles and markers of CVD risk in individuals with prediabetes and dyslipidaemia. † Deceased.

Technological advancements, unbalanced nutrition, sedentary life style, are important factors in obesity. Obesity-inflammation relationis beingexamined. In this study, the relationships among new generation inflammatory markers in children with normal body mass index (C) as well as obese (OB), morbid obese (MO) children and those with metabolic syndrome (MetS) were investigated. Materials and Methods: A total of 172 children participated in the study.Group 1 comprised children with normal body-mass index (control group) (C). Obese (OB) children were in Group 2, MO children constituted Group 3 and Group 4 included MO children with MetS. The number of cases were 37, 34, 51 and 50 in groups 1, 2, 3 and 4, respectively. Anthropometric measurements were recorded. Serum spexin, adropin, adipolipin, fibroblast growth factor-21 and fetuin-A levels were determined. Statistical analyses were performed. Result: Spexin and adipolin levels were significantly lower in obese groups than C group (p<0.05). Although adropin and FGF-21 levels did not differ significantly between groups, levels were lower in OB, MO, and MetS groups than C group.There were no significant differences among fetuin-A levels of the groups. Correlations between spexin and adipolin were the highest. These cytokines were negatively correlated with obesity parameters. The correlations between these cytokines were weakened from C group to MetS group. Conclusion: Decreasing spexin and adipolin levels in accordance with increasing obesity degrees and weakening of the correlation between these cytokines in MO group compared to C group may be helpful during the further investigation of obesity.
