Post-Translational Modifications of H2A Histone Variants and Their Role in Cancer

Abstract: Histone variants are chromatin components that replace replication-coupled histones in a fraction of nucleosomes and confer particular characteristics to chromatin. H2A variants represent the most numerous and diverse group among histone protein families. In the nucleosomal structure, H2A-H2B dimers can be removed and exchanged more easily than the stable H3-H4 core. The unstructured N-terminal histone tails of all histones, but also the C-terminal tails of H2A histones protrude out of the compact structure of… Show more

“…The altered expression of H2As has been found in cancers and resulted in different consequences. Whereas the H2AFX and H2AFY are considered as tumor suppressors, H2AFZ exhibits an oncogenic function [43]. Here, we further show that H2AFJ likely serves as a driver in promoting TMZ resistance and may be associated with the mesenchymal differentiation in GBM.…”

“…H2A.X was considered as exerting tumour suppressive roles and is considered as a biomarker for drug response [21]. In triple-negative breast cancer and melanoma, S139 phosphorylation of H2A.X was occurred and was correlated with poor prognosis [22][23][24].…”

“…The expression ratio of MacroH2A1.1 and MacroH2A1.2, the two isoforms of H2AFY, may be of great signi cance. MacroH2A1.1 acts as a tumour suppressor by inhibiting cell proliferation, migration, and invasion, while the function of MacroH2A1.2 is largely dependent on the type of cancer (18,32). In differentiated and proliferating cells, MacroH2A1.1 and MacroH2A1.2 have obvious expression differences: MacroH2A1.1 is primarily found in differentiated cells, while MacroH2A1.2 is mainly found in proliferating cells (33,34).…”
