“…Previous size estimates have been in the region of 2-5 kb (Smith & Harris, 1990), although Tacke et al (1990) obtained a value of 2.3 kb by locating the 5'-terminal nucleotide 40 residues upstream of the coat protein initiation codon. However, our results using a Scottish isolate of PLRV suggest a larger molecular size and a 5' origin further upstream.…”

“…This RNA is not encapsidated into viral particles (Smith & Harris, 1990) and contains the gene for the coat protein (CP, Tacke et al, 1989), which is separated by an amber stop codon from the ORF for a 56K protein (Fig. I a).…”

“…The ORF 3'-proximal to the NY-RPV coat protein ORF is proposed to be expressed by readthrough of the coat protein gene termination codon, as with a similar ORF of Vic-PAV and, indeed, with luteoviruses in general. A conserved 14 nucleotide sequence surrounding the luteovirus 22K coat protein gene termination codon has been identified previously, and a role for this sequence in the expression of the readthrough ORF postulated (Smith & Harris, 1990;Veidt et al, 1988). An examination of this region from NY-RPV indicates that this conserved region is actually the 12 nucleotide sequence AAAUAGGUAGAC, with the coat protein gene termination codon indicated by the underlined nucleotides.…”
