Potential anthelmintic effect of chitosan on Syphacia muris infecting Wistar rats: biochemical, immunological, and histopathological studies

Chitosan is a natural non-toxic, biocompatible, biodegradable, and mucoadhesive polymer. It also has a broad spectrum of applications such as agriculture, medical fields, cosmetics and food industries. In this investigation, chitosan nanoparticles were produced by an aqueous extract of Cympopogon citratus leaves as a reducing agent. According to the SEM and TEM micrographs, CNPs had a spherical shape, and size ranging from 8.08 to 12.01 nm. CNPs have a positively charged surface with a Zeta potential of + 26 mV. The crystalline feature of CNPs is determined by X-ray diffraction. There are many functional groups, including C꞊C, CH2-OH, C–O, C-S, N–H, CN, CH and OH were detected by FTIR analysis. As shown by the thermogravimetric study, CNPs have a high thermal stability. For the optimization of the green synthesis of CNPs, a Face centered central composite design (FCCCD) with 30 trials was used. The maximum yield of CNPs (13.99 mg CNPs/mL) was produced with chitosan concentration 1.5%, pH 4.5 at 40 °C, and incubation period of 30 min. The antifungal activity of CNPs was evaluated against phytopathogenic fungus; Fusarium culmorum. A 100% rate of mycelial growth inhibition was gained by the application of 20 mg CNPs/mL. The antitumor activity of the green synthesized CNPs was examined using 6 different cell lines, the viability of the cells reduced when the concentration of green synthesized CNPs increased, the IC50 dose of the green synthesized CNPs on the examined cell lines HePG-2, MCF-7, HCT-116, PC-3, Hela and WI-38 was 36.25 ± 2.3, 31.21 ± 2.2, 67.45 ± 3.5, 56.30 ± 3.3, 44.62 ± 2.6 and 74.90 ± 3.8; respectively.

Immune response, oxidative stress, and histological changes of Wistar rats after being administered with Parascaris equorum antigen

Worldwide, particularly in developing nations, helminth infections are the leading causes of livestock illness and mortality. Parascaris (P.) equorum, a parasitic worm from the Ascarididae family, significantly impacts the production, health, and working performance of equines. This study aimed to investigate the impact of intraperitoneal sensitization of P. equorum on the immune system, oxidative stress, and histology in Wistar rats. After acclimatization for 7 days, we divided the rats into five groups, each consisting of six rats. Group I, serving as the control, was administered distilled water, followed by groups II (day 7), III (day 14), IV (day 21), and V (day 33). The rats were euthanized every day mentioned (Days 7–33). On day 0, a dosage of 1ml/100 gm rat (containing 500 μg/ml protein content) emulsified crude antigen extract with an incomplete Freund’s adjuvant (1:1 volume), followed by a second dose of the same antigen concentration on day 7. To assess the allergenicity of this nematode, we measured a whole blood profile, serum levels of IFN-γ, IL-5, IL-10, IL-13, and IL-33, total immunoglobulins IgE and IgG, and oxidative stress markers. Also, we examined histological changes in the liver, kidney, and spleen. The results showed that values of total leukocyte count, granulocytes, monocytes, and lymphocytes were significantly (P < 0.05) increased on day 14 post-infection relative to other days of investigation. It was found that the levels of total immunoglobulins (IgE and IgG) and cytokines (INF-γ, IL-5, IL-13, and IL-33) on days 14 and 21 were significantly higher than in the control group. At all periods of the experiment, the injected group exhibited significantly higher concentrations of MDA and NO compared to the control group (P < 0.05). Conversely, GSH and CAT levels (P < 0.05) dropped significantly on days 7, 14, and 21. Different rat tissues showed alterations. Ultimately, this study described the detrimental effects of P. equorum crude antigen administration on the immune system, oxidative states, and histological changes of Wistar rats at various intervals.

An innovative approach to control fish-borne zoonotic metacercarial infections in aquaculture by utilizing nanoparticles

Fish-borne zoonotic trematodes (FBZTs) pose significant health risks and economic challenges worldwide. This study investigated the prevalence of encysted metacercariae (EMCs) in Nile tilapia (Oreochromis niloticus) from two Egyptian governorates and evaluated the antiparasitic efficacy of chitosan, silver, and selenium nanoparticles against these parasites. A cross-sectional analysis of 453 O. niloticus specimens revealed an overall EMC prevalence of 40.8%, with infection rates of 34.11% in Giza and 49.5% in Dakahlia. Clinostomid and Prohemistomid metacercariae were the most common, with mixed infections observed. Transmission electron microscopy characterized the synthesized nanoparticles, showing average diameters of 9.6–18.7 nm for chitosan, 13.2–19.8 nm for selenium, and 11.7–15.1 nm for silver nanoparticles. In vitro antiparasitic assays demonstrated varying efficacies among the nanoparticles. Against Clinostomum spp. metacercariae, chitosan nanoparticles showed the highest potency, achieving LC50 at 66 μg/ml after 30 min and LC90 at 100 μg/ml after 120 min. For Prohemistomum vivax EMCs, chitosan nanoparticles exhibited superior efficacy, achieving LC50 at 8 μg/ml after 1 h and LC90 at 16 μg/ml after 2 h. Silver and selenium nanoparticles showed lower efficacy for both parasite species. Scanning electron microscopy revealed significant ultrastructural damage to the parasite tegument following nanoparticle exposure, including disappearance of transverse ridges, integument shrinkage, and formation of blebs. This study provided valuable insights into the prevalence of FBZTs in Egyptian Nile tilapia and demonstrated the potential of nanoparticles, particularly chitosan, as effective antiparasitic agents. These findings pave the way for developing novel, targeted strategies to control fish-borne zoonotic trematodes, potentially reducing their impact on public health and aquaculture economies.
