Background: Metabolic associated fatty liver disease (MAFLD) affects a quarter of the worldwide population, but no drug therapies have yet been developed. Croton urucurana Baill. (Euphorbiaceae) is a medicinal species, that is, widely distributed in Brazil. It is used in popular medicine to treat gastrointestinal, cardiovascular, and endocrine system diseases. However, its hepatoprotective and lipid-lowering effects have not yet been scientifically investigated.Aim of the study: The present study investigated the effects of an extract of C. urucurana in a rat model of MAFLD that was associated with multiple risk factors, including hypertension, smoking, and dyslipidemia.Material and Methods: The phytochemical composition of C. urucurana was evaluated by liquid chromatography-mass spectrometry. Spontaneously hypertensive rats received a 0.5% cholesterol-enriched diet and were exposed to cigarette smoke (9 cigarettes/day for 10 weeks). During the last 5 weeks, the animals were orally treated with vehicle (negative control [C-] group), C. urucurana extract (30, 100, and 300 mg/kg), or simvastatin + enalapril (two standard reference drugs that are commonly used to treat dyslipidemia and hypertension, respectively). One group of rats that were not exposed to these risk factors was also evaluated (basal group). Blood was collected for the analysis of cholesterol, triglyceride, alanine aminotransferase (ALT), and aspartate aminotransferase (AST) levels. The liver and feces were collected for lipid quantification. The liver was also processed for antioxidant and histopathological analysis.Results: The main constituents of the C. urucurana extract were flavonoids, glycosides, and alkaloids. The model successfully induced MAFLD, reflected by increases in AST and ALT levels, and induced oxidative stress in the C- group. Treatment with the C. urucurana extract (300 mg/kg) and simvastatin + enalapril decreased plasma and hepatic lipid levels. In contrast to simvastatin + enalapril treatment, C. urucurana reduced AST and ALT levels. Massive lesions were observed in the liver in the C- group, which were reversed by treatment with the C. urucurana extract (300 mg/kg).Conclusion:C. urucurana extract exerted promising hepatoprotective and lipid-lowering effects in a preclinical rat model of MAFLD.

Plants used in traditional medicine may constitute an important source of new biologically active compounds. Acalypha indica is one of the weed plants that contain important medicinal values for human health applications. The present study was undertaken to assess the hepatoprotective activity of methanolic leaf extract of Acalypha indica in goat liver slice culture against carbon tetrachloride (CCl4) to prove its efficacy against liver disorders CCl4 was used to induce hepatotoxicity in liver slice of goat. The cytotoxicity induced by CCl4 was estimated by quantifying the release of marker enzymes such as alanine transaminase (ALT), aspartate transaminase (AST), alkaline phosphatase (ALP), acid phosphatase (ACP) and lactate dehydrogenase (LDH). Also, the degree of hepatic damage was measured by estimating the levels of lipid peroxidation (LPO) of hepatocytes membrane lipids. The treatment of liver cells with CCl4 caused twice increase in LPO of cells besides release of LDH, ALT, AST, ACP and ALP by 6.55, 3.89, 3.31, 2.69 and 2.50 times, respectively as compared to untreated liver cells. Thus, a toxic effect of CCl4 was significantly reduced by the treatment of methanolic leaf extract of Acalypha indica, silymarin and quercetin. The free radical scavenging activity of plant extract was estimated by 2, 2-diphenylpicrylhydrazyl (DPPH) method and IC50 is 65.12 ± 2.27 μg/mL. Qualitative analysis of flavonoid indicates the presence of flavonoids in plant extract. These results indicate that all concentration of plant extract of Acalypha indica, silymarin and quercetin protect the liver cells from CCl4 induced oxidative/free radical mediated damage in vitro. The hepatoprotective activity of methanolic extract of Acalypha indica extract is due to the presence of flavonoids which have remarkable antioxidant and anti-inflammatory activity. Cancer is one of the most common human diseases and a leading cause of human mortality in rencent years. Doxorubicin (DOX) is an important anti-cancer agent belonging to anthracyclines drug category. This chemotherapeutic agent acts against cancer cells through different mechanisms; however, its use is related with numerous acute and chronic dose-related side effects and toxicities including hepatotoxicity. The aim of this study was to conduct a comprehensive review of in vitro, in vivo and any human studies regarding the protective effects of synthetic compounds against DOX-induced liver injury.The search was conducted in embase, PubMed and Scopus databases using the following keywords: “doxorubicin”, “Adriamycin”, “hepatotoxicity”, “liver injury”, “liver damage” and “hepatoprotective” to find experimental and preclinical studies regarding the effects of various compounds on DOX-induced hepatotoxicity. Twenty-one eligible studies to the end of May 2022 finally were reviewed (19 in vivo and 2 both in vivo and in vitro studies). Our results demonstrated that all of these drugs, except acetylsalicylic acid, had considerable hepatoprotective effects against DOX (deferoxamine had a mild effect). In addition, except glutathione, none of the studied drugs and compounds attenuated the antitumor efficacy of DOX treatment. Moreover, no human study was available in this fieild. Altogether, our results demonstrated that most of compounds with anti-apoptosis, anti-inflammatory and antioxidant properties are efficacious against DOX-induced hepatotoxicity and may have potential for being considered for inclusion in the chemotherapeutic regimes of cancer patients, after fully been assessed in well-desgined large clinical trials.
