Potential Role of Macrophages in Experimental Keratomycosis

Abstract: Local depletion of macrophages may downregulate the immune response and aggravate fungal keratitis. Macrophages appear to play an important role in host defense against corneal infection of F. solani and C. albicans.

“…In line with the previous observation that TLR4 is expressed on intracorneally infiltrating macrophages during fungal keratitis, 28 we observed that a substantial proportion of F4/80-positive macrophages expressed TLR4 in injured cornea on day 2 postinjury (Supplemental Figure IIA). HMGB1 protein was detected in macrophages at the same time (Supplemental Figure IIA).…”

“…[31][32][33][34][35] Given the fact that TLR4 is expressed on macrophages to mediate inflammatory responses, 28 we examined ϩ macrophages started to accumulate on day 1 and reached a peak on day 4 after injury ( Figure 2A and 2B). Although the kinetics of macrophage infiltration in the wounds were similar in control and TLR4 Ϫ/Ϫ animals ( Figure 2B), the infiltration of macrophages in TLR4 Ϫ/Ϫ wounded corneas was significantly attenuated ( Figure 2B).…”

“…(Hu et al, 2009) In vitro studies have shown that inactivated hyphae of Fusarium solani upregulate gene expression of TLRs-2, 3, 4 and 6, along with protein expression of TLRs 2 and 4 with resultant increases in cytokine expression of IL-6 and IL-8. The importance of TLR-4 in modulating the host defense mechanisms to fungi, especially Fusarium keratitis, has been demonstrated by several groups.…”
