Potential viral vectors for the stimulation of mucosal antibody responses against enteric viral antigens in pigs

Abstract: Four viruses were compared for their ability to induce an intestinal antibody response in piglets. Antibodies were not detected in response to oral vaccination with either fowlpox virus or a baculovirus (BV). Simultaneous oral dosing and parenteral inoculation with high concentrations of ~v in an oil emulsion adjuvant induced high levels of circulating virus neutralising (VN) antibodies, and also low levels of intestinal antibodies when booster doses of virus were given. In response to oral vaccination with sw… Show more

“…Therefore, they may be particularly well suited as recombinant live vectors (Tuboly et al, 1993) for antigen delivery at the respiratory mucosal lymphoid tissues. Human adenovirus type 5 (Ad5), the serotype most commonly used in current work, can accept inserts of up to 4 kbp of foreign DNA following a partial deletion of the non-essential early transcription region 3 (E3) from the genome (Graham & Prevec, 1991).…”

“…All piglets were weighed and slaughtered at week 10. Digesta from three different parts of the small intestine were collected as described by Tuboly et al. (1993) and tested for the presence of TGEV and secretory (s)IgA antibodies.…”

“…The idea of using PAdV (genus Mastadenovirus, species Porcine A, B, C) vectors was published over 20 years ago (Tuboly et al, 1993). A few years later it was discovered that the virus can enter but is unable to replicate in dog, sheep, bovine, and human cells (Reddy et al, 1999a).…”
