“…Amino acids were recovered quantitatively in the eluate from the resin washed with five bed volumes of ammonia (2 N) (18). The amino acid fraction was dried by rotary film evaporation and excess ammonia was removed from the residue while it was stored in the presence of concentrated H2SO4 at reduced pressure for 8 h. The residue was dissolved in water to give 0.05 to 0.5 mM amino acids and these were converted to the corresponding 5-dimethylaminonaphthalene-l-sulfonamino acids (DNS-amino acids) using DNS-Cl as previously described (24) except that the volumes were reduced to one-tenth and the reaction was not terminated with an amine.
