Predicting the efficacy of radiotherapy in individual glioblastoma patientsin vivo:a mathematical modeling approach

Abstract: Glioblastoma multiforme (GBM) is the most malignant form of primary brain tumors known as gliomas. They proliferate and invade extensively and yield short life expectancies despite aggressive treatment. Response to treatment is usually measured in terms of survival of groups of patients treated similarly but this statistical approach misses the subgroups that may have responded to or may have been injured by treatment. Such statistics offer scant reassurance to individual patients who have suffered through the… Show more

“…Radiotherapy is a major component of treating glioblastoma. 25,26 MEK1/2 and CHK1 inhibitor treatment radiosensitized GBM cells either when irradiation occurred with drug exposure (concomitant) or shortly after drug exposure (Tables 1 and 2). The results are similar to what have been observed in non-CNS tumor cell types.…”

“…This work demonstrates that the response to carbon ions can be predicted from the response to photons and that ceramide plays an important role in the final phase of radiation-induced mitotic catastrophe regardless of p53 status. This type of archive is intended to facilitate customization of treatment for patients by allowing the development of new predictive mathematical models [26,27] for the response of tumors to radiation and ultimately improve hadrontherapy treatment plans.…”

“…the pioneering paper of Bahrami and Kim [1] and also the book of Wheldon [42] for early survey). Most available papers on mathematical modeling of gliomas have considered specific aspects of radiation therapy for high-grade gliomas [2,5,12,13,31]. As to LGGs there have been very few relevant works using mathematical models to describe the response to RT.…”
