Prediction of functional profiles of gut microbiota from 16S rRNA metagenomic data provides a more robust evaluation of gut dysbiosis occurring in Japanese type 2 diabetic patients

Abstract: We assessed whether gut microbial functional profiles predicted from 16S rRNA metagenomics differed in Japanese type 2 diabetic patients. A total of 22 Japanese subjects were recruited from our outpatient clinic in an observational study. Fecal samples were obtained from 12 control and 10 type 2 diabetic subjects. 16S rRNA metagenomic data were generated and functional profiles predicted using “Phylogenetic Investigation of Communities by Reconstruction of Unobserved States” software. We measured the parameter… Show more

“…16S rRNA sequencing using a MiSeq TM system (Illumina, San Diego, CA) was conducted according to a previously described method. 15) PCR amplification of 16S rRNA V3-V4 region was conducted using primers 314F and 805R. Processing of sequencing data including quality filtering, chimera check, OTU definition, and taxonomy assignment was carried out using QIIME, 16) USEARCH, 17) and UCHIME 18) exactly as previously described.…”

The relative abundance of bacterial genus between groups was analyzed by White's non-parametric t-test, which is a method developed to compare bacterial abundance.…”

“…Intestinal dysbiosis has been demonstrated to be associated with inflammatory bowel disease (IBD) [3,4], irritable bowel syndrome [5], and nonsteroidal anti-inflammatory drugs-induced enteropathy [6]. Moreover, gut dysbiosis has been reported in patients with types 1 and 2 diabetes [7,8], multiple sclerosis [9], autism [10], and rheumatoid arthritis [11]. …”
