Predictive impact of circulating microRNA‐193a‐5p on early relapse after autologous stem cell transplantation in patients with multiple myeloma

Abstract: We explored prognostic roles of circulating microRNAs (miRs) in multiple myeloma (MM) treated with autologous stem cell transplantation (ASCT) following induction chemotherapy. In part I of the study (n = 40), we identified a decreasing dynamics of circulating miR-193a-5p expression from diagnosis to pre-ASCT. In patients who experienced early relapse within one year post ASCT (n = 9) these patterns were distinctive compared to those without early relapse in a 1:2 matched cohort (n = 18). In part II (n = 90), … Show more

“…Low circulating levels of let-7b and miRNA-18a have been signi cantly associated with poor overall survival as well as progression free survival 36 and were found to be downregulated in this composite analysis too. Similarly, higher circulating miR-193a-5p has been shown to be an important prognostic biomarker for reduced probability of early relapse and better PFS following autologous SCT 37 . On the contrary, down regulation of circulating miRNA-193 has been associated with favoured progression of cancer in osteosarcoma and colorectal cancer 38,39 .…”

“…In preclinical studies, melphalan sensitivity of MM cells was inversely proportional to miR-221/222 expression (43). In the presence or absence of human bone marrow stromal cells, Up to day, only several studies have evaluated the role of miRNAs in MM patients undergoing AHSCT (59)(60)(61)(62)(63)(64)(65)(66)(67). However, differences in study designs, including sample collection timing and miRNA evaluation methods, make direct comparisons of results challenging.…”

“…Occasionally, busulfan (3.2 mg/kg/day for three days), melphalan (70 mg/m 2 /day for two days), busulfan (3.2 mg/kg/day for three days), melphalan (100 mg/day)-thiotepa (150 mg/m 2 /day for one day), and busulfan (3.2 mg/kg/day for three days)-thiotepa (150 mg/m 2 /day for one day) were used when melphalan was unavailable, as described in a previous report. 19 Some patients received an experimental regimen of bortezomib, busulfan, and melphalan in phase II clinical trials. 20 Other supportive care, including prophylactic antibiotics, prophylaxis for hepatic sinusoidal obstruction syndrome, granulocyte colony stimulation factor, and bisphosphonates, was administered concurrently across the three centers, as described in a previous report.…”
