Preferential radiosensitization to glioblastoma cancer stem cell‑like cells by a Hsp90 inhibitor, N‑vinylpyrrolidone‑AUY922

Abstract: The present study examined the radiosensitization induced by a heat shock protein 90 inhibitor, N-vinylpyrrolidone (NVP)-AUY922, in CD133-positive cells in a hypoxic area of T98G spheroids. CD133-positive cells that are induced in the hypoxic microenvironment of spheroids have previously been reported to exhibit cancer stem cell-like properties. The present study used CD133-positive cells from a glioblastoma cell line (T98G) as cancer stem cell-like cells. CD133-positive and negative cells were sorted from T98… Show more

“…Despite the opposing findings on the effect of Hsp90 on the survival of patients, experimental studies show that inhibition of the Hsp90 ATPase domain with Shepherdin, a peptidomimetic antagonist of the complex between the chaperone and survivin, resulted in induction of autophagy in glioblastoma cells and suppression of tumor growth in mice model (immunocompromised CB17 SCID/beige female mice), as well as longer survival [ 99 ]. Moreover, inhibition of Hsp90 in glioblastoma cancer stem cell-like cells (T98G) increased their sensitization to the radiotherapy [ 100 ]. Clearly, inhibition of Hsp90 has shown a positive effect on survival in vivo and in vitro [ 101 , 102 ].…”

“…The spheroids were then embedded in an embedding medium (Tissue-Tek ® O.C.T. Compound, Sakura Finetechnical Co., Ltd., Tokyo, Japan) and sliced into frozen sections 10–20 µm in thickness using a cryostat (Cryostar NX50™, Thermo Fisher Scientific Inc., Carlsbad, CA) ( 25 ).…”

“…The human glioblastoma cell line, T98G (JCRB, Tokyo, Japan), was used in this study. T98G cells were cultured in α-minimum essential medium (α-MEM) supplemented with 20 mM 4-(2hydroxyethyl) piperazine ethane sulfonic acid, 8 mM NaHCO 3 , 50 μg/ml streptomycin, 50 U/ml penicillin, and 10% fetal bovine serum, and maintained in a humidified incubator at 37°C under conditions of 98% air and 2% CO 2 (23)(24)(25).…”

“…Inhibitors of Hsp90 (e.g., geldanamycin, 17DMAG, radicicol) have been shown to enhance the radiosensitivity of various cell lines ( Table 1 ) [ 161 , 162 ]. Tani and colleagues found N -vinylpyrrolidone (NVP)-AUY922 enhanced radiosensitivity in CD133-positive GBM cells [ 163 ].…”
