PreliminaryIn VitroEvaluation of Genistein Chemopreventive Capacity as a Result of Esterification and Cyclodextrin Encapsulation

Abstract: The present study focuses on the synthesis and analysis of a genistein ester derivative with myristic acid followed by beta cyclodextrin encapsulation; physicochemical analysis using consecrated techniques such as FTIR, MS, DSC, and SEM revealed both a successful esterification and inclusion inside the cyclodextrin cavity. Cytotoxic effects were measured in vitro on three human cell lines: HeLa (cervix adenocarcinoma), A2780 (ovary carcinoma), and A431 (skin epidermoid carcinoma). The in vitro biological analy… Show more

“…Melanoma cells, specifically Skmel-103, exhibited a high sensitivity to GN, as expected due to their higher metabolic activity compared to 3T3 cells, which are non-malignant. This heightened sensitivity to GN in melanoma cells has been previously reported in various studies (Danciu et al, 2015;Dobrzynska, Napierala, Florek, 2020;Ji et al, 2012;Rauth, Kichina, Green, 1997;Tyagi, Song, De, 2019;Venza et al, 2018).…”

“…As per Zhang et al (2015), GN typically exhibits an endothermic peak at 301.7°C, which we observed in our study at around 303.7°C. Danciu et al (2015) also reported similar findings, with GN displaying an endothermic peak at 303°C, followed by an exothermic peak at 307°C, signifying chemical decomposition. In our DSC analysis of PNTPGS-GN (Figure 2-A) and PNPol-GN (Figure 2-B) formulations, it was not detected any additional peaks, confirming that no new composites formed due to degradation, chemical reactions, or incompatibilities.…”

“…The protocol used to obtain the cyclodetrins' complex, known as the kneading procedure, was already described in our previous scientific papers [10][11][12]: the active substance and hydroxypropyl-gamma-cyclodextrin -molar ratio 1:2 -were triturated with a mortar and a pestle, followed by kneading with a 50% ethanol solution until an important part of the solvent was evaporated. In the last step, the product was dried at room temperature for 24 h and then at 105 o C in a laboratory oven for several hours, until constant weight.…”

“…The new formulations did not decrease the viability of the cancerous cells. This behavior may be explained by the increased stability of the complex within the in vitro environment [138].…”
