Preliminary screening and identification of stem cell-like sphere clones in a gallbladder cancer cell line GBC-SD

Abstract: Abstract:This paper aims to screen and identify sphere clone cells with characteristics similar to cancer stem cells in human gallbladder cancer cell line GBC-SD. GBC-SD cells were cultured in a serum-free culture medium with different concentrations of the chemotherapeutic drug cisplatin for generating sphere clones. The mRNA expressions of stem cell-related genes CD133, OCT-4, Nanog, and drug resistance genes ABCG2 and MDR-1 in sphere clones were detected by quantitative real-time polymerase chain reaction (… Show more

“…Some investigators use cell surface markers to get the CSCs, such as CD44 + /CD24 −/low /Lin − for breast cancer CSCs and CD44 + /CD24 + for gastric CSCs (Al-Hajj et al, 2003;Zhang et al, 2011). Some developed tumor sphere in serum-free medium to obtain CSCs (Song et al, 2011;Yin et al, 2011). We selected FACS to obtain the cancer stem-like cells.…”

“…Putative CSCs in gallbladder cancer were identified with CD44 + CD133 + and CD133 + , respectively, as sorting markers [33,34]. Yin et al [35] obtained gallbladder CSC-like cells using suspension cultures of GBC-SD cells in serum-free culture medium containing CDDP. In the present study, we used SP cells as a model of CSC-like cells.…”

“…In contrast, the remaining five cell lines seem to represent stable in vitro models of distinct CSC-associated expression profiles. While we used adherent monolayer cultures following manufacturer recommendations, we can, however, not exclude that CSC-like expression profiles might be more variable and/or different if the cell lines analyzed here are processed as sphere 3D cultures [41,42,43,44]. …”
