Preparation and evaluation of anti-SARS coronavirus IgY from yolks of immunized SPF chickens

Abstract: Severe acute respiratory syndrome (SARS) is a recently discovered viral disease, characterized by fever, cough, acute fibrinous pneumonia and high infectivity. Specific pathogen-free (SPF) chickens were immunized with inactivated SARS coronavirus and their eggs were harvested at regular intervals. Yolk immunoglobulin (IgY) was extracted using the water dilution method, followed by further purification on a Sephadex G-75 column. SDS-polyacrylamide gel electrophoresis (SDS-PAGE), Western blot and neutralization … Show more

“…It could be lyophilized, reconstituted without loss of activity, and maintain good thermal stability. 143 Other theoretic treatments include treatment of SARS ARDS by blocking the pulmonary renin-angiotensin system or treatment with ACE-2. 37,38 Uncontrolled trials suggest that IFN alfacon-1 (a synthetic interferon) with steroids, protease inhibitors with ribavirin, or convalescent plasma with neutralizing antibody may be useful for treatment.…”

“…However, chicken egg is also considered as a major source of antibodies. In the past few years, many kinds of specific antibodies have been yielded successfully from eggs of immunised hens, which include anti-HIV-1 and anti-SARS coronavirus antibodies (Devi, Bai, Lal, Umashankar, & Krishnan, 2002;Fu et al, 2006;McLaren, Prosser, Grieve, & Brissenko, 1994;Ruan, Ma, He, Meng, & Zhu, 2005;Soler, Barquet, & Lucio, 1998). Presently, IgY has been used extensively for prevention and treatment of various infections in animals and humans with mixed success (Hamada & Kodama, 1996).…”

“…The oral administration of specific IgYs has been shown to be an alternative method for the treatment and prevention of gastrointestinal pathogens including Salmonella SPP. (Chalghoumi et al, 2009;Pavic et al, 2012), Escherichia coli (Sunwoo et al, 2006), Bovine coronavirus (Fu et al, 2006), Bovine rotavirus (Vega et al, 2011), Pseudomonas Staphylococcus (Wang et al, 2011).…”
