Preparation and in vivo evaluation of apatite/collagen packed composite by alternate immersion method and Newton press

Abstract: Further development of bio-compatible, bio-absorbable, and osteo-conductive bio-materials is desired for bone grafts in dental and medical clinics. One candidate material might be a high-density apatite/collagen composite, which cures relatively large bone defects. To produce such a composite, we freeze-dried type I collagen solution, cross-linked the formed sponge by 2 wt % glutaraldehyde, immersed the insoluble sponge in CaCl(2) and Na(2)HPO(4) solutions alternately five times, and compacted the sponge by Ne… Show more

“…Col itself does not have strong osteoconductive properties ( 28 ). In brief, we prepared Col, n-HAP/Col and m-HAP/Col specimens, and conducted an expression analyses of four osteogenic differentiation marker genes in SaOS-2 cells cultured for 1, 2, 3 and 4 weeks on three test materials (Col, n-HAP/Col and m-HAP/Col) by RT-PCR.…”

“…Various biomimetic methods are available to fabricate scaffolds for bone tissue. These methods include (i) mimicking the chemical composition of bone tissue (biopolymers and calcium phosphates nanocomposites), (ii) imitating hierarchical bone structure in micro- and nanoscales (delicate arrangement of collagen nanofibers, which are reinforced by hydroxyapatite nanoparticles), and (iii) developing manufacturing procedures, which replicate nature’s actual process of bone nanocomposite formation. Previously, we generated hydroxyapatite scaffolds for applications in bone tissue engineering. The main focus of those studies was mimicking the calcium phosphate part of the bone, which supported cell attachment and growth. In the current study, we mimic the structure of the bone concerning the organic and inorganic phases and evaluate the efficacy of fabricated scaffolds in vivo .…”
