Objective: Enalapril maleate (EM) is an angiotensin-converting enzyme (ACE) inhibitor. It is generally prescribed for the treatment of hypertension, heart failure and chronic kidney diseases in adults and children. EM 2.5 mg, 5 mg, 10 mg and 20 mg tablets and EM injection 1.25 mg/ml are currently available in the market. But a liquid paediatric formulation of this medicine is not currently available. Also, it is a BCS class III drug, having a bioavailability of approximately 60%. The present study proposes a new strategy for improvement of drug bioavailability and taste masking: EM nanoencapsulation within polymeric nanoparticles suspensions prepared with Eudragit RS100 (ERS100) as polymer and Tween 80 as a stabilizer, aiming at obtaining a liquid dosage form suitable for paediatric administration.Methods: Nanoprecipitation method used for the preparation of nanoparticles suspension. The preparations were evaluated for drug content, entrapment efficiency, particle size, zeta potential, polydispersity index, pH, viscosity and in vitro drug release. Based on the entrapment efficiency, viscosity and in vitro drug release the optimized formulation was selected. Optimized formulation evaluated for taste, ex vivo intestinal permeation, differential scanning calorimetry, scanning electron microscopy and release kinetic studies.Results: The optimized nanoparticle formulation F8 having drug to polymer ratio of 1:100 showed satisfactory drug content (95.1%), entrapment efficiency (77.71%), particle size (198.47 nm), pH (6.36), viscosity (2.9 x〖10〗^(-3)Pa•s) and 81.2% drug release after 12 h. The formulation has taste-masking properties and shows 84.6% drug permeation through the goat intestine within 12 h.Conclusion: The prepared nanoparticles suspension of Enalapril maleate was found to be an effective liquid pharmaceutical dosage form for paediatric administration with taste-masking properties.

Objective: The study aimed to develop a transetosome system as a delivery system of 4-n-Butylresorcinol (4nBR) and evaluate their physicochemical characteristics and skin penetration capacity compared with another vesicles system. Methods: Transethosomes were prepared through cold methods and the optimization of the formulation was carried out using “Box–Behnken design” approach from Design-Expert software (version 13.0. 3. 0, State-Ease Inc., Minneapolis, MN). The independent variables were soya lecithin, surfactant (Tween 80: Span 80 with a ratio of 1: 3) and Ethanol. The prepared formulations were characterized for vesicle size, polydispersity (PDI), zeta potential using a particle size analyzer and entrapment efficiency. Furthermore, transethosomes were formulated in serum preparations that tested for in vitro penetration test compared to serum with ethosomes, transfersomes and non-vesicles system. Results: Transethosomes formula optimization using box benhken approach produced a formula of 5.53 % soya lecithin, 3 % surfactant (Tween 80: Span 80 with a ratio of 1: 3) and 30 % Ethanol. The optimized formulation obtained particle size result of 197.4 nm; Polydispersity Index 0.421; zeta potential-56.8 mV and entrapment efficiency 98.40 %. Transethosomes serum met physical stability tests and in vitro penetration test showed better results compared to serum with ethosomes, transfersomes and non-vesicles system; the percentage of cumulative penetrated amounts of transethosomes serum, transfersomes, ethosomes and non-vesicle serum, respectively, was 41.43%; 23.59%, 19.85% and 2.43%. Conclusion: Development of 4nBR transethosomes using surfactant as edge activators and ethanol as an enhancers through optimization with box Behnken design resulted in transethosomes composition as ultra-deformable vesicles that fulfiled the physical characteristics, stability and permeability of 4nBR.

Formulation and Evaluation of Simvastatin Nanoparticles

Present study was aimed to formulation and evaluation of Simvastatin nanoparticles. Simvastatin nanoparticles were prepared by High pressure homogenization method by using lipid (Glyceryl Monostearate), polymer (Chitosan, Sodium Alginate) and surfactant (Tween 80) in the formulation. The prepared nanoparticles were evaluated for particle size, zetapotential, entrapment efficiency, drug excipients compatability studies by FTIR, scanning electron microscopy and invitro release studies. Among all the formulations, The formulation F8 showed particle size 97.47nm, PDI as 0.333, zeta potential -30.60mV and % Entrapment efficiency 85.69%, FTIR studies shown that drug was compatable with excipients, Scanning electron microscopy results shown that particles in the formulation shows moderately spherical in shape and drug release maximum 97.28%. So, formulations F8 showed better sustain the drug release for 12hrs period of time as compared to other prepared formulations, hence drug release kinetics was applied for F8 formulation. Drug release kinetic studies it showed that optimized formulation F8 showed zero order with non-fickian mechanism.
