Preparation and properties of a drug release membrane of mitomycin C with N-succinyl-hydroxyethyl chitosan

Abstract: A novel drug loaded membrane made of N-succinyl-hydroxyethyl chitosan and mitomycin C was used as an implant for glaucoma filtering surgery. The characteristics of the membrane, such as FTIR, equilibrium water content, swelling ratio, permeability, and drug release in vitro were determined. The L929 fibroblast inhibition of drug loaded membranes was compared to hydroxyethyl chitosan film and blank control, detecting by MTT. The biodegradability and biocompatibility were evaluated by implanting membranes into t… Show more

“…On the other hand, the chitosan cross-linked with GA membranes and the sample neutralized and cross-linked with GA shows a swelling degree of 1000 % after 72 h in PBS solution (equation 2) which is quite high when compared to the swelling index of ~100 % found by Sangsanoh [6] or 600 % found by Li et al [28]. Knault et al suggests that the swelling of chitosan films is mainly influenced by ionic interactions between polymer chains, and depends on the cross-linking density achieved during the formation of the biopolymer network.…”

“…[ 23 25 ] It is valuable for the drug carrier to readily prepare its conjugates with various drugs to avoid vexatious complications. [ 26 ] Theoretically, a chemical combination could happen between PTX and SHEC in the presence of an appropriate catalyst. Then, the membrane of PTX-SHEC may be prepared using cross-linked modification.…”

“…The absorption bands were characterized and compared with other MMC FTIR analyses. 30,34,40 Broadband at 3,448 cm -1 was observed, which refers to the characteristic absorption peaks of the combination of OH, NH, and NH 2 groups. Peaks at 2,860 and 2,926 cm -1 correspond to the CH aromatic band, and the absorption bands at 1,000 to 1,250 cm -1 were assigned to C-O and Si-O.…”

“…Several methods have been investigated for the delivery of MMC in nano or micro size. Common methods for MMC delivery consist of encapsulation of the drug in nano-or micro-particles, using biodegradable polymers including albumin, 27 dextran, 28 estradiol, 29 N-succinyl-chitosan, 30 hydrogels, 31 polybutylcyanoacrylate, 32 poly-epsilon-caprolactone, 33 oxidized-dextran microspheres, 28 and polylactic acids (PLA) nanoparticles. 34 However, to our knowledge, MMC has never been loaded into pSi.…”
