Preparation, characterization and biocompatibility of calcium peroxide-loaded polycaprolactone microparticles

Abstract: 目的 探究聚己内酯(PCL)包载过氧化钙(CPO)微粒的理化特征及生物相容性。 方法 制备CPO/PCL微粒,并采用扫描电子显微镜和能谱仪分别观察并比较CPO、PCL和CPO/PCL三种微粒的微观形貌特点和元素分布情况。提取大鼠脂肪间充质干细胞并加入不同浓度的CPO和CPO/PCL微粒(0.10%、0.25%、0.50%、1.00%),检测各组微粒的氧气和过氧化氢释放量。在缺氧/常氧环境下,加入以上不同浓度CPO/PCL微粒,给予细胞正常培养或成骨分化培养,实时逆转录聚合酶链反应检测碱性磷酸酶(ALP)、Runt相关转录因子2(RUNX2)、骨桥蛋白(OPN)和骨钙素(OCN)基因表达。随后,将1.00% CPO/PCL微粒注射于SD大鼠皮下,于7、14 d时采用苏木精-伊红染色(HE染色)和免疫组织化学染色观察微粒注射后组织的病理学变化和免疫细胞变化情况。 结果 CPO微粒为多边形结构,PCL微粒呈小球状塑料粒子的状态,CPO/PCL微粒为块状晶体结构。能谱仪下CPO微粒出现明… Show more
