Preparation, evaluation and metabolites study in rats of novel amentoflavone-loaded TPGS/soluplus mixed nanomicelles

Abstract: Amentoflavone (AMF) is a kind of biflavonoids existing in Ginkgo biloba leaves. It has many biological activities, such as antioxidant, anti-inflammatory, anti-bacterial, antiviral, hypoglycemic, anti-tumor and inducing apoptosis. However, its solubility and bioavailability are poor and there are a few studies on it in vivo. In this study, to improve its solubility and bioavailability, the nanomicelles were prepared with TPGS and soluplus as carriers for the first time. The particle size, Zeta potential, encap… Show more

“…The solubility of TH in different surfactant mediums was evaluated to satisfy the sink conditions [13]. Briefly, excessive TH was added to 10 mL of different medium at 37.0 ± 0.5 • C, the suspensions were under vigorously shaking for 48 h. The suspension was centrifuged at 1500× g and the pellet was discarded.…”

“…Curcumin however, requires piperine to enhance uptake 104 and amentoflavone may not have adequate uptake from source plant species such as Gingko biloba. 105 Our treatment array fit these requirements. A limitation of the strategy is that at the present time, we do not have precise estimations of bioavailability of individual compounds and bioavailability under conditions of combined uptake when taken together.…”

“…The difficulty of solubilizing higher concentrations of the compound in a vehicle that can be applied subcutaneously was a challenge. The difficulty in dissolving amentoflavone in water and organic solvents is known ( Feng et al., 2020 ). In order to improve solubility in liquid solvents, there are several pharmacotechnical possibilities.…”

“…In order to improve solubility in liquid solvents, there are several pharmacotechnical possibilities. Recently, micelles and nanomicelles have been used to improve the solubility and bioavailability of amentoflavone ( Zhang et al., 2019 ; Feng et al., 2020 ). Also, the use of amorphous solid dispersion contributed to the increase in solubility, dissolution and oral bioavailability of amentoflavone, promoting the antitumor effect in rats ( Chen et al., 2020 ).…”
