Preparation of human hyperimmune globulin to Haemophilus influenzae b, Streptococcus pneumoniae, and Neisseria meningitidis

Abstract: As a first step in exploring the feasibility of passive antibody prophylaxis and therapy of serious infections caused by common encapsulated bacteria, we have immunized healthy adults with Haemophilus influenzae type b vaccine, 14-valent pneumococcal vaccine, and meningococcal group A and C vaccine; collected plasma by repeated pheresis; and purified a hyperimmune globulin termed bacterial polysaccharide immune globulin by the cold-ethanol fractionation method of Cohn and Oncley. Specific antibacterial antibod… Show more

“…Serum from a normal non-vaccinated adult was included in each ELISA run to control for interassay variability. Antibody concentrations in patient samples were expressed relative to a reference adult hyperimmune plasma pool [24]. This plasma pool contains 2648 ng antibody N/ml for serotype 3; 1196 ng antibody N/ml for serotype 4; 539 ng antibody N/ml for group 6; 927 ng antibody N/ml for group 9 and 440 ng antibody N/ml for group 19 [24].…”

“…The reaction was stopped with 100 /il 2 N H1SO4, Optical density was measured on a Titertek FLISA reader at 450 nm. An anti-PRP hyperimmune serum (containing 32,5 /ig specific antibody ml [25]) was used as a standard. The standard was assigned arbitrarily to a level of lOO",.…”

“…However, the field was not completely abandoned for meningococcal disease, although greatly restricted to experimental research and isolated clinical experiences. [143][144][145][146][147] Passive antibody therapy for infectious diseases then became limited to infections not treated with antibiotics, such as the treatment and prevention of hepatitis B virus, rabies virus, respiratory syncytial virus, Clostridium tetani, Clostridium botulinum, anthrax, Clostridioides difficile colitis, vaccinia virus, echovirus, and enterovirus. 148 However, looming threats in the field of infectious diseases have made the scientific community turn its sight back to the use of antibodies for therapeutic use.…”
