Development and validation of stir bar sorptive extraction of polar phenols in water followed by HPLC separation in poly(vinylpyrrolididonedivinylbenzene) monolith An effective and simple method for polar phenols in water matrix was developed by using stir bar sorptive extraction (SBSE) based on a hydrophilic poly(vinylpyrrolididone-divinylbenzene) (VPDB) monolithic material and HPLC analysis. To achieve optimum extraction performance for phenols, several parameters, including extraction and desorption time, desorption solvent, pH value, and ionic strength of sample matrix, were investigated. Under the optimized experimental conditions, eight phenols were directly enriched from water samples and analyzed by HPLC-DAD. The detection limits (S/N = 3) and quantification limits (S/N = 10) of the proposed method for the target compounds were achieved within the range of 0.72 -1.37 and 2.40 -4.27 ng/mL from spiked water, respectively. Recoveries of eight phenolic compounds were found in the range of 55.2 -95.9%. The calibration curves showed the linearity ranging from 5 to 150 ng/mL with linear regression coefficient R 2 values above 0.98. Method repeatability presented as intra-and interday precisions were also found with the RSDs less than 4.10 and 7.61%, respectively. The distribution coefficients between VPDB and water (K VPDB/W ) for phenolic compounds were also calculated and compared with K O/W . Finally, the proposed method was successfully applied to the determination of the target compounds in tap water, sea water and wastewater samples.

Recebido em 3/8/07; aceito em 17/12/07; publicado na web em 4/9/08 STIR-BAR SORPTIVE EXTRACTION FOR DRUGS ANALYSIS IN BIOLOGICAL FLUIDS. A novel solventless sample preparation, stir-bar sorptive extraction (SBSE), for extraction, and sample enrichment of organic compounds from biological fluids, is described in this manuscript from principle to applications. The SBSE is based on sorptive extraction, whereby the compounds are extracted into a polymer coating, polydimethylsiloxane (PDMS), on a magnetic stirring rod. The extraction is controlled by the partitioning coefficient of drugs between the PDMS and sample matrix, and upon the sample-extraction medium phase ratio. The SBSE technique has been applied successfully, with high sensitivities, to biomedical analysis of volatiles and for semi-volatiles drugs from biological sample, including urine, plasma, and saliva. SBSE combined with in situ derivatization, drugs quite more polar (e.g. metabolites) also can be analyzed.Keywords: stir-bar sorptive extraction (SBSE); drugs; biological fluids. introduçãoEm razão da complexidade dos fluidos biológicos e da baixa concentração dos fármacos nestas matrizes, a etapa de preparo de amostra -extração, pré-concentração dos analitos e eliminação dos interferentes -tem sido requerida no desenvolvimento de métodos cromatográficos com alta sensibilidade e seletividade analítica.Os métodos convencionais, empregados no preparo de amostras biológicas, para análises cromatográficas, têm sido a extração líquido-líquido e extração em fase sólida. [1][2][3][4][5][6] A miniaturização dos sistemas analíticos tem sido uma convergência predominante na área de química analítica. A microextração em fase sólida (SPME -solid-phase micro-extraction), in-tube SPME e a extração sortiva em barra de agitação (SBSE) são exemplos típicos de técnicas de preparo de amostra miniaturizadas. Estas técnicas, quando hifenadas a sistemas analíticos de detecção, têm resultado em análises rápidas, baixo consumo de solvente, menor exposição dos analistas aos fluidos biológicos e aos solventes tóxicos, assim como maior precisão analítica e automação das análises. 7A microextração em fase sólida apresenta uma série de vantagens em relação aos métodos de preparo de amostras convencionais, ou seja: não requer instrumentação analítica sofisticada, não utiliza solvente orgânico, rápido processo operacional, permite automação das análises, a reutilização das fibras extratoras e integra em um único sistema, a extração, concentração e introdução da amostra no sistema cromatográfico. 8,9 Porém, em razão do pequeno volume das fases extratoras e limitado número de fibras SPME, seletivas para análises de fármacos, os métodos analíticos padronizados têm apresentado baixa recuperação analítica, tornando necessária a utilização de sistemas de detecção de alta sensibilidade.Em 1999, Baltussen e colaboradores 10 apresentaram a técnica extração sortiva em barra de agitação, utilizando uma barra de agitação magnética revestida com polidimetilsiloxano (PDMS), a fase extratora.Os v...
