Preparative Purification of Anti-Proliferative Diarylheptanoids from Betula platyphylla by High-Speed Counter-Current Chromatography

Abstract: Abstract:A simple and rapid method using high-speed counter-current chromatography (HSCCC), along with bioassay-guided fractionation based on the anti-proliferative activity against renal and colon cancer cells, has been developed for the preparative separation of aceroside VIII (1) and platyphylloside (2) from Betula platyphylla. A solvent system composed of ethyl acetate/acetonitrile/water (1:0.1:1, v/v/v) was optimized for the separation. The upper phase was used as the stationary phase, and the lower phase… Show more

“…Thus, peaks 1 and 13, sugar and fumaroprotocetraric acid isomer 1, respectively, were present in all MAEs, whereas fumaroprotocetraric acid isomer 2 (peak 16) was found in Italia and Filipina MAE. Moreover, aceroside VIII (peak 42) was tentatively identified for the first time in M. alba , which was found in plants from the same class [ 25 ]. Finally, peak 54 which showed a m / z at 555.2819 was only found in the Valenciana Temprana genotype and was also detected in botanicals from the same order [ 26 ].…”

“…In this study, we employed solid tumor and blood cancer cell lines to evaluate the potential of BPP as a multitarget therapeutic agent, in both colon cancer and leukemia. To evaluate the mechanism of BPP in vitro, a large amount of BPP (Figure 1A) was isolated from the CH 2 Cl 2 fraction of the B. platyphylla bark extract using high-speed counter-current chromatography, as previously described [20]. In terms of the structure–activity relationships, in vitro results from previous studies suggest that the structure of BPP, which consist of two aromatic rings joined by a seven carbon chain along with a glucosyl group at C-5 and a carbonyl group at C-3, positively correlated with anticancer activities [20,23].…”

“…Betula platyphylla bark was obtained from the afforested land of SK E & C (Seoul, Korea), and a voucher specimen (SNU-797) was deposited in the Herbarium of the Medicinal Plant Garden, College of Pharmacy, Seoul National University, Korea. High yield platyphylloside (5.0%) was isolated from the Betula platyphylla CH 2 Cl 2 fraction using high-speed counter-current chromatography, as previously descried [20].…”
