Presence and distribution of biogenic amines (histamine, serotonin and epinephrine) in immunophenotyped human immune cells

Abstract: Human lymphocytes contain the three biogenic amine, similar to rat. However, while each amine was present in monocytes, in granulocytes serotonin and epinephrine were not demonstrated. The results call attention to the possible extrapolation of animal data to human lymphocytes and monocytes, but in the case of granulocytes, caution is needed. Taking into consideration earlier results, activated T cells appear to have an important role in the loss or production of hormones inside the immune system.

“…This supports the role of the H4R in AD, where particularly in the chronic phase, lower levels of histamine are released from infiltrating immune cells [9]. It has been shown that histamine is released after de novo synthesis in various cells involved in the inflammatory response, such as monocytes, eosinophils, and T cells [10]. Furthermore, coexpression of the H1R, H2R, and H4R on diverse immune cells has been detected.…”

“…Prior to isolation of the spleen, the C57BL/6 mice were pretreated with chloro-DLphenylalanine methyl ester hydrochloride (pCPA, 250 mg/kg) to diminish endogenous store of 5-HT and therefore to diminish effects due to intrinsic 5-HT [11,20,21]. IL-2 and IFN-γ production from the purified splenic CD4 or CD8 T cells that were stimulated with Con A was then measured in the presence or absence of the selective 5-HT2AR agonist, DOI, or its antagonist, sarpogrelate.…”

“…SP induces proinflammatory cytokine secretion from inflammatory cells, which amplifies local inflammation [31,32]. SP produced by the nervous system can influence the function of the stomach, and the behavior of the immune cells and the materials (proinflammatory cytokines, chemokines, SP, hormones) produced by lymphocytes and mast cells themselves can each influence the production of the other materials [33,34]. Elevated SP level is postulated to be a significant pathophysiological factor in the development of gastritis.…”
