Presence and distribution of leucocyte subsets in the murine epididymis after vasectomy

Abstract: Male mice were vasectomized by 'open-ended' or 'closed' techniques. After 4 weeks the cell-mediated immune reactions were compared with those of sham-operated animals by immunohistochemical localization of leucocytes, using specific monoclonal antibodies. Macrophages and MHC class II antigen-positive cells were the major cell types to appear in all regions of the epididymis after both types of operation. There was recruitment of T-helper/inducer leucocytes but not of T-suppressor-cytotoxic cells. An increased … Show more

“…These changes in vasectomized mice were considered indicative of chronic inflammation and increased antigen presentation (Nashan et al, 1990). Whereas definition of functional subsets of leukocytes by staining with antibodies has not yet been accomplished in vasectomized Lewis rats, it is notable that the present ultrastructural observations also identified features of chronic inflammation and revealed many opportunities for presentation of antigens to lymphocytes by macrophages.…”

“…We have directed attention to the occurrence of inflammatory lesions in epididymal interstitial tissue after vasectomy in the Lewis rat (Flickinger et al, 1990a), and postvasectomy epididymal inflammation has also been described recently in the mouse (Nashan et al, 1990). These reactions are characterized by accumulation of mononuclear cells in the connective tissue, and the presence of an interstitial reaction is correlated with higher serum antisperm antibody levels (Flickinger et al, 1990a).…”

“…The origin of leukocytes from the epididymis has been confirmed by studies performed using the vasectomy model,3637 and these models produce evidence of local inflammatory responses following the procedure. However, in patients with leukocytospermia, seminal citric acid concentrations (a marker of prostate function) appear to be lower, which suggests that prostatitis is the main cause of the presence of leukocytes in the semen.…”

“…Epididymal basal lamina was thickened, interstitial collagen became abundant and inflammatory lesions were observed in the tunica propria surrounding the tubules following vasectomy (Nashan et aL , 1990;Flickinger et al , 1990). In the present study, while there no epithelial alterations in the ipsilateral epididymis 3 months after vasectomy, collagen accumulation was seen in the interstitial tissue.…”
