Presence and secretion of atrial natriuretic peptide from cultured human aortic endothelial cells

Abstract: The endothelium is the production site of several potent vasoactive substances that modulate vascular tone and growth. The present study was undertaken to investigate the presence and secretion of atrial natriuretic peptide (ANP) immunoreactivity from vascular endothelial cells. ANP immunoreactivity was present in cultured human aortic endothelial cells by both immunohistochemical staining and radioimmunoassay. ANP immunoreactivity was also detectable in culture medium from human aortic endothelial cells in lo… Show more

“…ET-1 secretion is regulated predominantly by transcription and is modulated by various hormones and physical forces (11,13). Only small amounts of ET-1 are stored in vascular endothelial cells and most are released after synthesis (14,15). These studies exclude acid-induced decreased ET-1 degradation, increased release of preformed ET-1 from cellular compartments, and accelerated conversion of big ET-1 to ET-1 as mechanisms of increased ET-1 secretion caused by reduced extracellular pH.…”

“…Previous evidence indicates that ANP can be expressed and secreted by aortic endothelial cells [100], suggesting a local action of ANP also in the endothelium. Mice with endothelial-specific deletion of NPR-A gene (NPR-A-ecKO) undergoing limb ischemia displayed impaired angiogenesis until 5 weeks post-surgery.…”

“…ANP infusion also influences the sympathetic nervous system, probably by inhibiting the baroreceptormediated activation of renal sympathetic nerves as well as increasing noradrenaline release [405,414,415]. ANP causes antimitogenic and antiproliferative effects in glomerular mesangial cells, in vascular smooth muscle cells, and in the heart [416] and arterial wall [417]. ANP inhibits the synthesis of the potent vasoconstrictor endothelin-I [418] and induces apoptosis in neonatal rat cardiomyocytes [419].…”
