Presence of both mating types of Ascochyta rabiei in Argentina suggests potential for sexual reproduction

Abstract: The presence of both mating types was tested for in twelve isolates of Ascochyta rabiei in Argentina. A MATspecific PCR assay was performed. Nine of these isolates were identified as MAT1-1 and three of them MAT1-2. This is the first research regarding the mating types of Ascochyta rabiei in the country and suggests that there is a potential for sexual reproduction in Argentina.

“…In a work recently published by our research group, the presence of both mating types was analysed by multiplex-PCR in the 12 isolates obtained from different sites of Córdoba province (Crociara et al, 2020). In that work, we reported that nine of the isolates were MAT1-1 idiomorph and three were MAT1-2 idiomorph.…”

“…Plant tissue segments (10-mm long) and/or seeds were surface sterilized with ethanol 70% (30 s) and sodium hypochlorite 0.2% (2 min), and washed three to four times with sterile water. Each piece was then placed in Petri dishes containing sterile water agar supplemented with streptomycin (150 ml/L) and incubated at 21°C, under a 12/12 h fluorescent light/ dark cycle (Valetti et al, 2020). After 7 days, Petri dishes were visually examined for the presence of the typical structures described for A. rabiei: pycnidia, cirrus and conidia (Chen et al, 2011;Harveson et al, 2011).…”
