Presence of native limbal stromal cells increases the expansion efficiency of limbal stem/progenitor cells in culture

Abstract: Niche factors are important in the maintenance and regulation of stem cells. Limbal stromal cells are potentially a component of limbal stem cell (LSC) niche. We investigated the role of the limbal stromal cells in the ex vivo expansion of limbal stem/progenitor cells. Limbal epithelial cells were cultured as single-cell suspension and cell clusters from dispase II or collagenase A (ColA), or tissue explant. ColA isolated limbal stromal cells along with limbal epithelial cells. In the presence of limbal stroma… Show more

“…This finding is consistent with a previous observation [14]. LSCs cultured in the form of cell clusters achieve a higher expansion efficiency of the progenitor cell population [19, 20]. In contrast, LSC clusters cultured in the 2D system produced cell outgrowths with a very heterogeneous morphology and a significantly low number of p63α bright cells; the presence of large differentiated cells at the center of the outgrowth, away from the feeder cells, suggested that nutrients secreted by the BMSC feeder cells could not reach this central area.…”

“…A common disadvantage of the explant over the cell suspension method is the small amount of cells obtained because of the low proliferative rate in the former [5355]. Moreover, LSC isolated by cell suspension can help reduce contamination by other cell types, such as fibroblasts [52].…”

“…9,10 MSCs in the limbal stroma are more primitive than those in corneal stroma and support LESCs, demonstrating that these stromal stem cells constitute a component of the limbal-niche (unpublished data). Therefore, it is possible that the hyperreflective structures on IVCM represent the limbal stromal niche.…”

“…Recent studies using human cadaver tissue examined the cellular components of the anterior limbal stroma and their role in the maintenance of stemness in the basal epithelium. [9][10][11][12] However, the structure and function of the anterior limbal stroma has not been studied in vivo in normal individuals or patients with LSCD. Hence the former is the objective of the present study using IVCM.…”
