Pretreatment-Integration for Milk Protein Removal and Device-Facilitated Immunochromatographic Assay for 17 Items

Abstract: Accurate and comprehensive immunochromatographic assay (ICA) data are urgently required in the daily supervision of plants, schools, testing institutions, and law-enforcing departments. Through pretreatment-integration and device-facilitated operation, a quantitative ICA with high sensitivity and throughput was realized on the basis of a commercialized semi-quantitative ICA strip. Three pretreatment methods, namely, acid base, heavy metal salt, and organic solvent methods, have less than three steps. The pretr… Show more

“…To assess the reliability of the aptasensor, real food samples, specifically milk and raw beef, were procured from a local market. The milk was pretreated using the acid−base approach, 58 which involved adding acetic acid to promote protein coagulation in 1 mL of milk. After 3 min of vigorous vortexing, the milk protein was denatured, and the solution was centrifuged at 22 °C for 3 min at 5000 rpm to obtain the clear supernatant.…”

“…Furthermore, a protein pellet consisting mostly of caseins may be collectible at the bottom of the centrifuge vial. Protein separation is further promoted by mixing the milk with ammonium sulfate, an organic solvent, or an acid since these additives lead to the precipitation of milk proteins so that they can easily be removed by subsequent centrifugation or filtration [ 1 , 12 , 64 , 65 , 66 , 67 ].…”
