“…Nevertheless, the use of animal models to study the association between obesity and asthma has been shown to be of relevance , and environmental‐related factors (allergens and diet) are specially relevant for the obesity and asthma epidemics seen in Western countries . Moreover, experiments required to fully understand the cellular and molecular mechanisms must be performed in animals for ethical reasons, specially those testing pharmacological agents . However, when extrapolating data from mice to humans, care should be taken with anatomical differences in respiratory tract, types of adjuvants, chronicity of the disease, and age of animals (infants vs adults).…”

“…The in vivo expression of the cloned gene is driven by the eukaryotic promotor over much longer periods of time as compared to protein antigens [90]. The DNA vaccine thereby better primes the immune system to offer long term immune memory and potentially support tolerance induction via promoting Treg cell responses [91,92]. Moreover, the built-in CpG motifs in the DNA vaccine backbone activate toll-like receptor 9 (TLR9) that leads to the activation of MyD88-dependent signaling pathways which drive Th1-oriented immune responses [93].…”

“…Overall, the main data, shown in key review papers, highlighted the capacity of DNA plasmid encoding allergens to prevent or revert the Th2-biased parameters of the allergic response (as airway eosinophilia, specific IgE and Th2 cytokines as IL-4, IL-5) through the development of allergen-specific Th1 responses characterized by interferon (IFN)g release and induction of specific IgG antibodies [9,10,15]. Some studies evidenced as well the ability of DNA vaccines to activate Treg cells as well as IL-10/TGF-b but mainly when co-administrated with protein allergen [16][17][18] (Fig. 1).…”
