Prevention of hypertension and vascular changes by captopril treatment.

Abstract: Treatment of female spontaneously hypertensive rats (SHR) and control Wistar-Kyoto (WKY) rats with captopril was carried out by the addition of the drug in the drinking water throughout pregnancy and lactation and after weaning. At 28 weeks of age, average systolic blood pressure of treated SHR was 113±3 mm Hg, which was below that of control SHR (188±3 mm Hg) and WKY rats (124±3 mm Hg). Body weight and heart rate of the SHR were not affected by the treatment Tissue level of catecholamines was increased by cap… Show more

“…11,12 Studies from the group of Berecek suggested that these results could result from a decrease in AVP levels. 13,14 Similar studies have been performed by other laboratories using both ACE inhibitors, 15,16 and angiotensin receptor blockers (ARBs). 17,18 In our initial studies, we found that treatment of stroke-prone SHR with an ACE inhibitor from age 3-10 weeks resulted in a sustained suppression of blood pressure, whereas such an effect was not found with the vasodilator hydralazine.…”

“…Mutations in the fibrillin-1 gene lead to Marfan syndrome, while fibrillin-2 mutations lead to congenital contractural arachnodactyly (Lee et al, 1991b;Park et al, 1998;Chaudhry et al, 2001). Mice lacking the fibrillin-1 gene (fbn12/2) die within two weeks of birth from vascular and pulmonary complications, including ruptured aortic aneurysms, impaired breathing and diaphragmatic collapse.…”

“…Furthermore, enalapril treatment has been shown to correct such an abnormality in Ca 2ϩ handling in arteries of SHRs. 52 Treatment with ACE inhibitors may reduce the contractile response to agonists, 53 thereby mechanically augmenting the relaxation. However, the maximal active tension generated by 10 Ϫ5 mol/L NE did not differ among the four study groups (unpublished data).…”
