Primary and novel approaches for colon targeted drug delivery – A review

Abstract: The colon is a site where both local and systemic delivery of drugs can take place. Local delivery could, for example, allow topical treatment of inflammatory bowel disease. Treatment could be made more effective if it were possible for drugs to be targeted directly on the colon. Systemic side effects could also be reduced. Colon specific systems might also allow oral administration of peptide and protein drugs, which are normally inactivated in the upper parts of the gastrointestinal tract. Primary approaches… Show more

“…[3,4] There are various primary approaches for achieving colon targeting, including delivering of drug as a prod rug, by delaying the drug release, or by microbial way. [5,6] However, these approaches have many disadvantages or have some limitations that enforce over new colon developed system which are better than former ones. Colonic drug delivery is useful to improve treatment of disease depending on the diurnal rhythm such as asthma and arthritis.…”

“…The presence of different pH values along the gastrointestinal tract (GI tract) is a considerable challenge for drugs to reach the target sites accurately [5,6]. Many strategies related to colon-specific drug delivery were introduced; these strategies include the covalent linkage between drugs and polymers (prodrug), pH, time and microbial responsive strategies osmotic pressure-controlled drug delivery system [7][8][9] and the use of multiparticulate systems, such as microspheres and nanospheres [10].…”

“…Creation of suitable environment for the growth of colonic microorganisms, storage reservoir of faecal contents, and expulsion of the contents of the colon at an appropriate time and absorption of potassium and water from the lumen are the major functions of the human colon. About 2000ml of fluid enters the colon through the ileocecal valve from which more than 90% of the fluid is absorbed showing that the absorptive capacity of colon is very high [15][16][17] .…”
