Primary cell culture of meningothelial cells—a new model to study the arachnoid in glaucomatous optic neuropathy

Abstract: The technique described in this paper for the primary culture of meningothelial cells from the subarachnoid space of the optic nerve and using magnetic beads for the removal of fibroblasts is effective in obtaining a highly enriched meningothelial cell culture.

“…To analyse the phagocytic capacity of MECs, we evaluated the potential of these cells to engulf and take up large particles. To this end, human WHO grade I meningioma derived, hTERT immortalised Ben‐Men‐I cells were used as model for MECs as previously established (Puttmann et al, ; Xin et al, ). As shown in Figure A, MECs incubated with increasing amounts of fluorescently labelled carboxylate‐modified latex beads displayed increasing intracellular fluorescence.…”

“…The increase of CSF pressure leads to proliferation of meningothelial cells [281,282]. These cells participate in immunological processes in the CSF [283].…”

“…Primary meningothelial cells derived from porcine optic nerves (PMECs) [16] were incubated with apoptotic bodies derived from monocytic U-937 or neuron-like SH-SY5Y cells and the uptake of apoptotic bodies was analyzed by confocal microscopy (Figure  1) as well as flow cytometry (Additional file 1: Figure S1). Primary MECs were found to actively ingest apoptotic bodies derived from monocytic (Figure  1A) as well as neuronal-like cells (Figure  1B).…”

“…Primary porcine meningothelial cells (PMECs) were cultured as described previously by us [16]. Monocyte-like U-937 cells (Sigma-Aldrich, 85011440) were cultured in RPMI 1640 supplemented with 10% FCS, 2 mM L-glutamine, and 1 mM sodium pyruvate (Sigma-Aldrich).…”
