Pro-C5, a marker of true type V collagen formation and fibrillation, correlates with portal hypertension in patients with alcoholic cirrhosis

Abstract: Plasma Pro-C5 reflects liver hemodynamics, liver function, disease stage and clinically significant portal hypertension (PH). A multimarker model in combination with clinical scores predicted HVPG and separated clinical relevant HVPG thresholds. Plasma Pro-C5 may be suitable for the noninvasive evaluation of PH in patients with cirrhosis.

“…The serological biomarkers of interstitial matrix turnover were assessed by type 3, 5, and 6 collagen formation (PRO‐C3, N‐terminal propeptide of type 5 procollagen [PRO‐C5], and N‐terminal propeptide of type 6 procollagen [PRO‐C6]), and type 3 collagen degradation was assessed by matrix metallopeptidase 9 (C3M). Basement membrane formation was evaluated by type 4 collagen formation (P4NP7S) and type 4 collagen degradation mediated by matrix metallopeptidases (C4M2).…”

“…We used validated competitive ELISAs (Nordic Bioscience, Herlev, Denmark) to assess true formation of interstitial matrix collagen type III and V (PRO‐C3 and PRO‐C5) and degradation of interstitial matrix collagen type III and basement membrane type IV collagen (C3M and C4M) in serum samples from PSC to ulcerative colitis patients. All biomarkers were assessed in a blinded manner according to the manufacturer and samples were measured within the detection range.…”

“…The interstitial ECM in fibrosis mainly consists of fibril‐forming type I and type III, and minor quantities of type V collagen. Type V collagen regulates fibril size, whereas types I and III are mainly structural. The precursors of these collagens consist of a large central triple helical region, with N‐ and C‐terminal propeptides covalently linked to each end.…”
