Pro‐inflammatory effects of hydrogen sulphide on substance P in caerulein‐induced acute pancreatitis

Abstract: Hydrogen sulphide (H2S), a novel gasotransmitter, has been recognized to play an important role in inflammation. Cystathionine-γ-lyase (CSE) is a major H2S synthesizing enzyme in the cardiovascular system and DL-propargylglycine (PAG) is an irreversible inhibitor of CSE. Substance P (SP), a product of preprotachykinin-A (PPT-A) gene, is a well-known pro-inflammatory mediator which acts principally through the neurokinin-1 receptor (NK-1R). We have shown an association between H2S and SP in pulmonary inflammati… Show more

“…In the exocrine pancreas, H 2 S was initially demonstrated to be a pro-inflammatory factor involved in caerulein-induced pancreatitis (5,53). Later, however, anti-inflammatory effects were also reported (50,54).…”

“…To induce mild acute pancreatitis, we gave AQP12-KO mice and their WT littermates seven hourly intraperitoneal injections of the CCK-8 analog caerulein (50 g/kg body wt; Sigma-Aldrich) as previously described (1,2). Blood was collected at both 9 and 24 h after the first caerulein injection to examine the serum amylase and lipase levels.…”

“…In addition to its known effects in nociception, it also has proinflammatory roles [85], and absence or inhibition of this receptor is protective against ALI [86][87][88]. Hydrogen sulfide (H 2 S) has also recently been shown to be an important mediator of inflammatory responses and may induce the production or release of substance P [89,90]. Although H 2 S may be proinflammatory, in certain models of ALI, H 2 S production is decreased, and administration of the H 2 S donor sodium hydrosulfide decreased proinflammatory cytokines IL-6 and IL-8, and the resultant ALI [91].…”
