Pro‐oxidant effects of 7‐hydroperoxycholest‐5‐en‐3β‐ol on the copper‐initiated oxidation of low density lipoprotein

Abstract: In low density lipoproteins (LDL) supplemented with aged cholesterol and oxidized in the presence of Cu 2÷, an increase of the lipid oxidation parameters was observed compared with pure cholesterol-enriched LDL. A compound, identified as 7-hydroperoxycholesterol (7HPC), isolated from aged cholesterol and added to LDL, reproduced the above effects. The results indicate that the pro-oxidant effect of 7HPC is dependent on the hydroperoxy group since the corresponding alcohol derivative, 7a-hydroxycholesterol, had… Show more

“…In the procedures used in the present paper, the endogenous peroxide content of the isolated LDL is an important determinant of the oxidizability of LDL for the copper-mediated oxidation, whereas using AAPH the mechanism in less dependent on the lipid peroxides. Besides fatty acid peroxides [24,25], cholesterol peroxides potentiated the Cu2+-initiated LDL oxidation [26]. However, these latter compounds did not account for the effects observed with CS in the present study since TLC-repurifled CS was as pro-oxidant as stock CS.…”

“…[10] In the present study, we investigated the respective involvement of sulfur groups in the antioxidant activity of HSA. Influence and importance of targeted modifications of Cys and Met by N-ethylmaleimide (NEM) and chloramine T (CT) were examined on HSA antioxidant activity assessed by the coppermediated oxidation of human low density lipoproteins (LDL) [11] and by the free radical-induced blood hemolysis tests. “…At higher temperatures, copper can also react nonenzymatically with cholesterol (35,36) or arachidonate (37) in the presence of oxygen to produce hydroperoxide derivatives. To test whether the ␤-amyloid reaction product was the same as that produced by copper alone, we reacted cholesterol (0.1 g) with 10 mM CuCl 2 under 100% oxygen in tetrahydrofuran at 60°C overnight.…”
