Probiotic culture of the rotifer Brachionus plicatilis

One of the pathways for the entry of bacteria, both pathogenic and probiotic, into the larvae of fish hatcheries is via live prey. As a preventive measure against infections, live prey may be cultured, supplementing the food with probiotics. Some lactic acid bacteria (LAB) have been successfully used in the larviculture. In this study, the nutritional effect of seven terrestrial LAB has been studied regarding the growth of the rotifer Brachionus plicatilis. The cultures were carried out without partial renewal of the culture medium, feeding the rotifers on baker's yeast and adding some of the species of bacteria. In all cases, the addition of the bacteria increased both the specific maximum growth rate and the maximum density obtainable in the cultures. However, the best results were obtained with the addition of Lactococcus lactis spp lactis, Pediococcus acidilactici or Lactobacillus casei ssp. casei. The rates of growth obtained with the individual or joint addition of these three bacteria were 8-13 times greater than those obtained with the control cultures after 4-5 days' culture. In this study, a series of kinetic models have been applied (logistic modified−Gompertz, logistic−logistic and generalised logistic) which describe the experimental data, obtaining a set of parameters of biological significance which facilitate the optimisation of the use of these bacterial strains in the mass production of rotifers.

Biotechnology can be defined as any technology that involves living organisms or their derivatives. In applying this definition to rotifers, we focus on their contribution in culturing of early larval stages of marine fish. After almost four decades of marine fish culture in captivity, the success of this worldwide industry is still quite dependent on mass culture of the species Brachionus plicatilis and B. rotundiformis. In mass culture, the rotifers are continuously driven to reproduce at high rates, in relatively extreme environmental conditions of high population density and high loads of organic matter. Therefore, the success of mass cultures and future improvements in these systems relies on a close interaction between basic and applied studies of rotifers. In the present review, we will attempt to analyze why rotifers are suitable for early life stages of fish and to describe, in general, methodologies that have been devised for reliable supply of rotifers in large quantities. Problems associated with rotifer production, nutritional quality and effect on fish health and nutrition, will be discussed. Research on B. plicatilis and B. rotundiformis has increased enormously during the past three decades and these two species are the best-studied rotifers so far. While much of the research on these species is directed or devoted to the needs of aquaculture industry, they are also used as models for addressing basic biological questions, due to the relative ease of culture and their availability. Studies on feeding, pheromones, speciation in rotifers, the occurrence and putative hormones involved in sexual and asexual reproduction and production of resting eggs, are few examples of such studies. Rotifers will probably maintain their role as food organism for fish larvae, in spite of attempts to replace them with more accessible formulated food. Development of new culture methods that will improve the nutritional quality and production efficiency of rotifers may result in more diversified and flexible tasks for these organisms in aquaculture.

RESUMEN.Los laboratorios acuícolas altamente dependientes de Nannochloropsis sp. desarrollan mecanismos para su producción masiva, con el desafío de poder extraer altas concentraciones celulares. La floculación por agentes alcalinos es una alternativa que promete mayores ventajas, y el hidróxido de sodio puede ser el agente floculante más conveniente. Sin embargo, para lograr una eficiencia máxima de floculación son determinantes la cantidad del agente floculante, tiempo de exposición, concentración celular y tipo de microalga. Por lo anterior, se determinó la cantidad de NaOH 0,5N y el tiempo de exposición necesarios para lograr un pH óptimo de floculación en cosechas de Nannochloropsis sp. con concentraciones celulares mayores a 50x10 6 cél mL -1. Los resultados mostraron que con 17 mL de NaOH 0,5N por litro de cultivo de microalgas se puede lograr una eficiencia de floculación de 94,9% a pH 9,7 en menos de 60 min. Las células del sobrenadante (subproducto del proceso) fueron viables como inóculo de un nuevo cultivo de microalgas. El concentrado microalgal (355,7x10 6 cél mL ) obtenido fue adecuado como alimento para rotíferos. Por lo tanto, se recomienda el empleo de NaOH 0,5N como método simple, práctico y de bajo costo para producir concentrados de Nannochloropsis sp. con fines acuícolas. Therefore, employment of 0.5N NaOH is recommended as a simple, convenient and inexpensive method for producing Nannochloropsis sp. concentrates for aquaculture purposes.
