Production and characterization of extracellular carbohydrate polymer from Cyanothece sp. CCY 0110

Abstract: Cyanobacterial extracellular polymeric substances (EPS) are heteropolysaccharides that possess characteristics suitable for industrial applications, notably a high number of different monomers, strong anionic nature and high hydrophobicity. However, systematic studies that unveil the conditions influencing EPS synthesis and/or its characteristics are mandatory. In this work, Cyanothece sp. CCY 0110 was used as model organism. Our results revealed that this strain is among the most efficient EPS producers, and … Show more

“…CCY 0110 (Culture Collection of Yerseke, The Netherlands) was grown in 100 ml Erlenmeyer flasks containing ASNIII medium [18] supplemented with 1 M MOPS buffer (pH 7.0), at 30 ºC under a 12 h light (50 µE/m 2 /s)/12 h dark regimen with magnetic stirring (150 rpm). These conditions were previously identified as maximizing Cyanothece´s growth and EPS production [17].…”

“…The total carbohydrate content and the amount of carbohydrates released to the culture medium (RPS) were measured using the phenol-sulfuric A C C E P T E D M A N U S C R I P T ACCEPTED MANUSCRIPT 6 acid assay [21], as described previously [17]. All experiments were performed with three biological replicates and all measurements were performed in triplicate.…”

“…However, the interest in EPSs has increased considerably in recent years because these compounds are candidates for many commercial applications. Numerous types of exopolysaccharides have already been described (Castellane and Lemos 2007;Monteiro et al 2012;Mota et al 2013;Radchenkova et al 2013;Silvi et al 2013).…”

“…In general, the EPS production showed a positive linear effect with light intensity especially in the presence of combined nitrogen for different Nostoc species [5], Arthrospira platensis [6] and Cyanothece sp. CCY 0110 [7]. However, the different Nostoc species yielded contradictory results indicating that EPS production is affected both by nutritional and environmental parameters even to a species-specific level [8].…”
