Production and characterization of monoclonal antibodies for aflatoxin B1

Abstract: . Hybridomas that secreted antibodies for aflatoxin B1 were selected using two immunization protocols referred to as A and B. Protocol A is a standard immunization method and resulted in the selection of only two clones that produced monoclonal antibodies against aflatoxin B1. In protocol B a unique immunization schedule which resulted in the generation of 10 hybridomas is described. Of the 10, one antibody was highly specific to B1, four antibodies reacted equally strongly with B1, G1 and weakly with B2. Anot… Show more

“…The resulting monoclonal antibodies all show similar selectivities. The affinity to the aflatoxins usually follows the order AFB1 > AFG1 > AFB2 > AFG2 [10][11][12][13]. For this type of conjugate the cross-reactivity pattern is in accordance with the hapten structure shown in Figure 2.…”

“…Some sensitive antibodies were produced for the determination of AFB 1 in different agricultural products (Candlish et al 1987;Devi et al 1999;Lee 2004;Kolosova et al 2006;Wang and Liu 2006;Li et al 2009). In addition, some ELISA methods were developed for the determination of AFM 1 in dairy products (Magliulo et al 2005;Rosi et al 2007;Anfossi et al 2008;Pei et al 2009;Zhang et al 2009;Guan et al 2011).…”

“…The lateral flow strips are simple, rapid, and qualitative methods, while ELISA was a sensitive and semi-quantitative method. During the last two decades, several ELISA methods and other immunoassays were developed for the determination of AFB 1 or AFM 1 (Candlish et al 1987;Devi et al 1999;Magliulo et al 2005;Wang and Liu 2006;Anfossi et al 2008;Li et al 2009;He et al 2012;Puiu et al 2012;Jinap et al 2012;Rossi et al 2012). The aim of our study was generation of a broad-specificity MAb, enabling development of a sensitive and reliable immunoassay capable of detecting a broader range of aflatoxins.…”
