Production of a Monoclonal Antibody against an Epitope on HeLa Cells that Is the Functional Poliovirus Binding Site

Abstract: SUMMARYCell lines of primate origin carry receptors on their plasma membrane which are responsible for the specific binding of poliovirus. This paper describes the isolation and characterization of a monoclonal antibody reacting with the plasma membrane of HeLa ceils. The antibody (D171) was selected for its protection of HeLa cells against the cytopathic effect of poliovirus type 1. This protection was found to extend to all three viral serotypes, while the replication of five other viruses in HeLa cells was n… Show more

“…The role of DAF (CD55), vitronectin receptor (a v b 3 ) and PVR (CD155) in cell attachment and virus internalization was studied using CPE protection assays (Nobis et al, 1985). None of the blocking antibodies was able to prevent infection in RD cells.…”

“…Two of the groups isolated mAbs which blocked infection with all three serotypes ofpolio and no other enteroviruses as expected from previous kinetic binding studies (52,53). These mAbs recognized receptors only on poliovirussusceptible primate cell lines.…”

“…All experiments were performed at least twice and qualitatively similar data were obtained. 39 the first anti-PVR antibody, was submitted to the 1993 Human Leukocyte Typing V White Cell Differentiation Antigen workshop. In this workshop, laboratories throughout the world screened numerous cell types for expression of D171 reactivity.…”
