Production of biodegradable rods of polycaprolactone/olanzapine for potential application in treatment of Schizophrenia

Abstract: Olanzapine (OLZ) is a commonly antipsychotic used to treat schizophrenia, a mental illness, with fewer side effects when compared with other drugs of the same class. It is commercialized mainly as oral tablets and orally disintegrating tablets. However, this drug oral bioavailability is affected by the first pass effect and low solubility in water. This requires high daily doses that can cause more side effects and poor treatment compliance. Also, the frequency of frequency of tablet intake is commonly related… Show more

“…Recently, de Almeida et al. ( 2021 ) reported a hot-melt extrusion method for the production of PCL/OLZ implants. These implants contained lower drug loading than the one reported here (23% w/w), and drug crystals were found in the surface of the implants.…”

“…Different forms, such as films, scaffolds, matrices, membranes, and others, have been made. In this context, polymeric materials, due to their properties, have been studied (ALMEIDA et al,2021;MENDONÇA, et al, 2012) .…”

“…Biomaterial science is so interdisciplinary and allows the design of new materials. Therefore, studies concerning the incorporation of drugs into polymeric devices are increasing (ALMEIDA et al, 2021;da SILVA et al .2021;MENDONÇA, et al, 2013). Thus, the interest in comprehending the absorption process is elementary to modifying polymer device properties.…”
