Two soil isolates of Microsporum gypseum were studied for the production of extracellular proteases. Both the strains secreted protease on glucose-gelatin medium. The enzyme activity peaked on day 15 at 28°C. Asparagine repressed protease yield. Sugars caused catabolite repression of protease formation. Protease activities of both the isolates were significantly affected by incubation period, culture media and carbohydrates used. Both the strains grew on the skin bait and caused a gravimetrically measurable loss of the substrate. Despite less pronounced differences in the keratinase levels, great variations occured in the amount of keratin degraded by two isolates. Keratinase production as well as loss in substrate mass was better in glucose-lacking flasks than those containing the sugar. Although the rate of keratin degradation was independent of enzyme production, statistically positive correlations were recorded between loss in substrate mass: yielded dry mycelial weight and substrate degradation: keratinase levels.

In this study, we identified culturable microscopic fungi in soil from areas frequented by people (parks, allotments, and other green areas) in the city of Wrocław (Poland). In addition to general species analysis, attention was focused on keratin-degrading fungi. From 60 soil samples (12 study sites), we obtained 75 isolates of keratinophilic and keratinolytic fungi using the hair bait method, and 54 isolates of fungi were isolated on PDA (Potato Dextrose Agar) medium. Based on morphological and molecular analyses, a total of 37 strains were identified, classified into 2 phyla, 11 families, 17 genera, and 30 filamentous species. The mean values of the Shannon Diversity Index for both experimental variants ranged from 0.074 to 0.117. The most common species was the Penicillium genus, which accounted for 33.33% of all fungal species obtained in these studies. These fungi are common in both indoor and outdoor environments. However, particularly noteworthy in this study are the species belonging to the group of dermatophytes (Arthroderma uncinatum, Keratinophyton wagnerii, Nannizzia gypsea, and Paraphyton cookei), which may pose a real biological threat to humans and animals due to their well-known potential to cause dermatomycosis.

Summary The production of elastase, urease and sulphatase by the primary isolations as well as by subsequent transfer cultures of Epidermophyton floccosum was studied. The granular culture did not produce enzymes studied. Production of all of the enzymes was recorded by the time when pleomorphic areas appeared in the culture. In subsequent transfer cultures made separately from the pleomorphic and granular growth areas, the pleomorphic cultures were active anzyme producers while a minimal or no production could be recorded in the granular cultures. These findings show that the transformation of the granular growth to the pleomorphic one is connected to activation of production of elastase, urease and sulphatase by E. floccosum. Zusammenfassung Es wurde die Erzeugung von Elastase, Urease und Sulfatase von Epidermophyton floccosum bei Erstisolierungen sowie deren Subkulturen untersucht. Die granulär wachsende Pilzform entwickelte keine der untersuchten Enzyme. Hingegen wurde die Produktion aller dieser Enzyme zu der Zeit verzeichnet, als pleomorphe Wachstumszonen zwischen den granulären Wuchsformen erschienen. Von den pleomorphen und den granulären Wuchszonen wurden getrennt Subkulturen hergestellt. Die pleomorphen Wuchstypen waren aktive Enzymbildner, während von den granulären Typen nur minimal oder keine Enzyme produziert werden konnten. Diese Ergebnisse zeigten, daß mit der Umwandlung des granulären Wuchstyps zum pleomorphen die Förderung der Enzymbildung — Elastase, Urease und Sulfatase — bei Epidermophyton floccosum in Gang gesetzt wird.
