Productive Replication and Evolution of HIV-1 in Ferret Cells

Abstract: A rodent or other small animal model for HIV-1 has not been forthcoming, with the principal obstacles being species-specific restriction mechanisms and deficits in HIV-1 dependency factors. Some Carnivorans may harbor comparatively fewer impediments. For example, in contrast to mice, the domestic cat genome encodes essential nonreceptor HIV-1 dependency factors. All Feliformia species and at least one Caniformia species also lack a major lentiviral restriction mechanism (TRIM5␣/TRIMCyp proteins). Here we inves… Show more

“…1, compare lanes 7 and 8). Interestingly, Rev expression stimulated VLP production Ͼ10-fold in cat (CRFK) and quail (QT35) cell types, a finding consistent with prior reports of significant Rev activity in nonhuman cell types other than those derived from rodents (43,(58)(59)(60). However, although hCRM1 exerted no effect on Rev activity in cat cells, VLP production in the avian cell line was enhanced a further ϳ10-fold when Rev and hCRM1 were expressed together ( Fig.…”

“…The domestic dog, cat, and ferret lack functional antiviral TRIM5alpha and TRIMCyp genes (29)(30)(31), leaving them without a version of the important postentry capsid-targeting systems that restrict retroviruses in the cells of numerous primates (32)(33)(34)(35); there is similarly no known carnivore equivalent to the mouse Fv1 gene (36). The cat does have an effective APOBEC3 protein repertoire, which is antagonized by feline immunodeficiency virus (FIV) Vif and feline foamy virus Bet (37)(38)(39)(40)(41).…”

“…Human HEK 293T and HT1080 cells; Crandell feline kidney (CrFK) cells from Felis catus ; ferret alveolar epithelial cells (FtAEpC) and ferret kidney cells, each from Mustella putorius furo ( 69 ); owl-monkey kidney cells (OMK; ATCC CRL-1556); mouse 3T3 cells; and pteropid bat cell lines derived by Crameri et al ( 30 ) from Pteropus alecto kidney (paKiT01), brain (paBrH02), fetus (paFeSV40T), and lung (paLuT02) were maintained in Dulbecco’s modification of Eagle’s medium (DMEM) with 4.5 g/liter glucose; 10% fetal bovine serum; and 1× penicillin, streptomycin, and l -glutamine. Following transduction with Tsin IRESpuro HIV-1 vectors, P. alecto and CrFK cells were selected in 2 and 5 μg/ml puromycin, respectively.…”
