Progesterone Action in Endometrial Cancer, Endometriosis, Uterine Fibroids, and Breast Cancer

Abstract: Progesterone receptor (PR) mediates the actions of the ovarian steroid progesterone, which together with estradiol regulates gonadotropin secretion, prepares the endometrium for implantation, maintains pregnancy, and differentiates breast tissue. Separation of estrogen and progesterone actions in hormone-responsive tissues remains a challenge. Pathologies of the uterus and breast, including endometrial cancer, endometriosis, uterine fibroids, and breast cancer, are highly associated with estrogen, considered t… Show more

“…It was affirmed that menopause as a life event has no effect on quality of life but the presence and intensity of climacteric symptoms may have a negative impact on all the aspects of quality of life (Kim et al, 2013). Despite a submission that physical, social and psychological changes that occur during the menopausal stage of women have negative effects on their QoL (Teoman, Özcan, & Acar, 2004), there has been presumption that ethnic diversity may be responsible for differences in quality of life of individuals in different geographical locations (Heringa et al, 2015).…”

“…5,6 In contrast, an understanding of the actions of progestins in the development and progression of breast cancer is controversial. 7,8 In general, it is thought that progestins can both inhibit and stimulate proliferation of breast cancer cells. 9 The Women's Health Initiative Study firmly established that treatment with progestin in combination with estrogen (E2) for attenuating postmenopausal symptoms increases the risk of breast cancer, particularly TNBC.…”

“…Endometriotic tissue produces large quantities of progesterone and contains much lower levels of progesterone receptors than endometrium [34]. Progesterone, induces much lower levels of prolactin expression in endometriotic cells compared to endometrial stromal cells, suggesting that progesterone resistance may lead to endometriosis.…”

“…Progesterone resistance is increased by the low progesteronereceptor levels in endometriotic tissue. In endometriotic tissue, levels of progesterone receptor isoform B (PR-B) is undetectable, while that of the progesterone receptor isoform A (PR-A) isoform is markedly reduced, in endometriotic tissues [29,34].…”
