Progesterone receptor expression contributes to gemcitabine resistance at higher ECM stiffness in breast cancer cell lines

Abstract: Chemoresistance poses a great barrier to breast cancer treatment and is thought to correlate with increased matrix stiffness. We developed two-dimensional (2D) polyacrylamide (PAA) and three-dimensional (3D) alginate in vitro models of tissue stiffness that mimic the stiffness of normal breast and breast cancer. We then used these to compare cell viability in response to chemotherapeutic treatment. In both 2D and 3D we observed that breast cancer cell growth and size was increased at a higher stiffness corresp… Show more

“… 30 PR expression is beneficial to the resistance of BC cells to gemcitabine. 31 Then, we included hsa-miR-3646, hsa-miR-4741, hsa-miR-6730-3p, hsa-miR-6831-5p, hsa-miR-8485, nation, ER ratio and PR ratio for the multivariate logistic regression analysis and the development of the nomogram model. The nomogram model has potential predictive values and practical implications in BC.…”

“…The physics of cancer is an emerging field of oncology which highlights the mechanical properties of the tumor microenvironment (TME), focusing on the role of cell-ECM interactions in cancer disease and metastasis ( Burgos-Panadero, Lucantoni, et al, 2019 ; Grant et al, 2022 ; Kpeglo et al, 2022 ; Liu et al, 2022 ; Xiao et al, 2022 ). As glycoproteins interact with integrins and are involved in cell adhesion and migration pathways, molecules similar to VN, such as FN have been used in cancer mechanotransduction studies ( Missirlis and Spatz, 2014 ; Elosegui-Artola et al, 2016 ).…”
