Prognostic significance of BLK expression in R-CHOP treated diffuse large B-cell lymphoma

Abstract: Background: The aim of the present study was to evaluate the prognostic significance of B-cell lymphocyte kinase (BLK) expression for survival outcomes in diffuse large B-cell lymphoma (DLBCL) patients treated with R-CHOP.Methods: We retrospectively analyzed the medical records of 89 patients from two tertiary referral hospitals. The expression of BLK, SYK, and CDK1 were evaluated in a semiquantitative method using an H-score, and the proportions of BCL2 and C-MYC were evaluated. Results: A total of 89 patients… Show more

“…Only few studies have, however, investigated longitudinal abundance of (EV-derived) cfRNA markers (87,88). When comparing pre-to interim-, and to posttherapy samples in DLBCL responders, a clear decrease in cell-free mRNA of B-cell related markers was noted, many of which have been proposed as potential diagnostic and/or prognostic biomarkers in DLBCL tissue (89)(90)(91)(92)(93)(94)(95)(96)(97)(98). Moreover, the interim timepoint showed a clear, transient increase in neutrophil-and inflammation-related markers, reflecting an acute-phase reaction during therapy.…”

“…Tissue sections from the corresponding formalin-fixed, paraffin-embedded blocks were mounted onto SuperFrost Plus Microscope Slides (Thermo Fisher Scientific, Waltham, MA, USA), deparaffinized in xylene, and then rehydrated using an ethanol gradient. Immunohistochemical staining was carried out using a Bond-Max automated immunostainer (Leica Biosystems, Lincolnshire, IL, USA) and a Bond Polymer Refine Detection System (Leica Biosystems), as described previously (11,30,(35)(36)(37)(38)(39). Following antigen retrieval, the slides were incubated with a primary antibody against L1CAM (dilution 1:100, clone EPR18998, Abcam, Waltham, MA, USA).…”
